Effects Of Shendi Bushen Capsule On The Expression Of PI3K/AKT Pathway In Rats With Renal Interstitial Fibrosis

Objective To observe the effect of Shendi Bushen capsule on expression of renal interstitial fibrosis and expression of PI3K/AKT signal transduction pathway in UUO rats through animal experiments,and to explore the mechanism of Shendi Bushen capsule on anti-renal interstitial fibrosis.Chinese medicine compound prescription provides theoretical basis for the treatment of renal interstitial fibrosis.Methods A total of 70 clean male wistar rats weighing 180-220 g were selected randomly.The rats were randomly divided into blank control group,sham operation group,UUO model group,Shendi Bushen group and urinary toxin group.The renal interstitial fibrosis model was made by unilateral ureteral ligation.After the model was successfully prepared,the rats were weighed daily at 1 ml/100 mg to the control group,sham operation group,and model group.The suspension prepared by the ratio of 0.324 g/kg/d was intragastrically administered;the rats in the uremic villus-treated group were intragastrically administered at a ratio of 1.81 g/kg/d and were intragastrically administered for 28 days.The general state of rats in each group was observed.Changes of serum creatinine,urea nitrogen,and urinary albumin were measured on the 14 th and 28 th day after administration.The degree of renal interstitial fibrosis was observed by HE staining and immunohistochemistry was performed.The method was used to detect the expression of PI3K/AKT pathway in the kidney.Results The pathological changes of Shendi Bushen group were lighter than that of uremic Qing group and model group at all time points.On the 14 th and 28 th days after modeling,the blood creatinine,blood urea nitrogen,and urinary microalbumin in Shenqi Bushen group were significantly reduced.Compared with the model group(P<0.05),there was statistical significance,and the treatment effect was better than the uremic clear group;on the 28 th day after modeling,the expression of PI3 K and AKT in the kidney of the Shendibu kidney group was significantly increased,compared with the model group.(P<0.05),statistically significant.Conclusion Shendi Bushen Capsule can significantly reduce the serum creatinine and blood urea nitrogen value of UUO rats and protect the residual renal function;ShendiBushen Capsule can reduce the loss of urinary microalbumin in UUO rats;Shendi Bushen Capsule may reduce the UUO The expression of murine PI3K/AKT signaling pathway delays the progression of renal interstitial fibrosis.
