Objective:To investigate the mechanism of renal interstitial fibrosis after unilateal ureteral obstruction and the renoprotective effect of Shentong capsule.Methods:Twenty-four rats were randomly assigned to sham operation group (normal group), operation group and Shentong treatment group after unilateral ureteral obstruction.Renal tissues were examined by light microscopy 8 days after operation. Renal fibrosis was assessed by measuring tissue hydroxyproline.Immunhistochemistry was applied to measured the expression of TGF- β1, α -SMA and Smad3,7, and the level of plasmic and renal Endothelin (ET-1) was measured by radioimmuoassay.Results: In comparison with sham operation group, the hydroxyproline of renal tissues in operation group significantly increased, the expression of TGF- β 1 , α -SMA and Smad3 significantly increased, and the expression of Smad7 significantly decreased, the level of plasmic and renal ET-1 significantly increased (P<0.01) , In comparison with operation group, the level of renal tissue hydroxyproline signicantly decreased in Shentong treatment group, Shentong capsule significantly decreased the expression of TGF- β 1, α -SMA and Smad3 and significantly increased the expression of Smad7, plasmic and renal ET-1 signicantly decreased in Shentong treatment group (P<0.05) .Conclusion: Shentong capsule may inhibit over the expression of TGF- 3 1 by inhibiting over the expression of Smad3 and promoting expression of Smad7,and inhibit over the expression of a -SMA and reduce the level of plasmic and renal ET- 1 ,thereby inhibit renal interstitial fibrosis.
