Background and objectiveTumor,as one of serve disease,which threaten human health and affected the quality of human life,It is a key project that the clinical researchers have been exploring,and the tumor is divided into benign tumors and malignant tumors,among them,malignant tumors are particularly affecting human life.Lymphoma is a malignant tumor originating in the lymph nodes or out of the lymph nodes,and is one of the most common malignant tumors in children and young people.The World Health Organization(WHO)classification defines each type of lymphoma with independent histopathological features,immunophenotypic,molecular genetics,and clinical manifestations.The lymphomas are divided into two major categories: Hodgkinlymphoma and non-Hodgkin lymphomas.There are more than 60 types of non-hodgkin's lymphoma in the 2016 updated WHO classification of hematopoietic and lymphoid tissue tumor,which is the most complicated classification of all tumors.Extranodal NK/T cell lymphoma(ENKTCL)is a rare and invasive non-hodgkin lymphoma subtype.It has potential relationship to Epstein-Barr virus(EBV)infection,this type of lymphoma has strong invasiveness and poor prognosis,mainly in Asia and Latin America,and it is one of the most common subtypes of lymphoma in our country.For traditional anthracycline chemotherapy,compared with other types of lymphoma,Extranodal NK/T cell lymphoma is resistant to treatment.At present,the standardized treatment of Extranodal NK/T cell lymphoma has not been recognized by international scholars.Because of its malignancy,invasiveness and threat to human life.Therefore,it is of great significance to study the mechanism of occurrence and development of extranodal NK/T cell lymphoma,related prognostic indicators or to find new therapeutic targets.MicroRNA(miRNA)is a small non-coding RNA composed of 18 to 25 nucleotides,it works through several different mechanisms,and the target gene can be negatively regulated after transcriptional level,they play an important biological role in regulating cell growth,development,apoptosis,invasion,metastasis and hematopoiesis.miRNA regulates the target gene expression by blocking the translation of mRNA or regulating the degradation of mRNA.The types and expressions of miRNA of each type tumer were different,so mi RNA can be used as markers of target cells.Therefore,the detection and analysis of the types and expressions of miRNA in tumer cells can be used as an important indicator for the treatment and prognosis of lymphoma.MiR-21-5p is processed by the 5 'end arm of the miR-21 precursor,human miR-21 gene is located in chromosome 17q23.The development of tumor involves many changes in miRNA,which involves mi R-21-5p,the expression level of miRNA is correlated with the infiltration and metastasis of tumer cells.For example,studies have shown that high-expressed miR-21-5p can regulate the survival,infiltration and metastasis of tumer cells in non-small cell carcinoma;Similarly,low expression of miR-21-5p can reduce the expression level of the related proteins in tumer cells.MiR-150-5p is processed by the 5 'end arm of the precursors of miR-150.According to the relevant research evidence,miR-150-5p plays an important role in the development of some tumors,including lymphohematopoietic tumors.MiR-150 family can regulate the expression level of FOXP1 and GAB1 protein in malignant and normal B cells,and the BCR signaling pathway can also be affected.Researches have shown that it is believed to have a role in promoting the proliferation of gastric cancer cells,and is thought to have overexpression of over 50 times the expression in osteosarcoma.According to published literature,miR-21-5p and miR-150-5p play an important role in tumors.At present,there is no specific study on miR-21-5p and miR-150-5p of Extranodal NK/T cell lymphoma in the relevant literature.Therefore,miR-21-5p and miR-150-5p were applied in the study of Extranodal NK/T cell lymphoma in this topic.Using real-time fluorescence Quantitative PCR(Quantitative real-time PCR,qRT-PCR)to detect the expression of miR-21-5p and miR-150-5p in Extranodal NK/T cell lymphoma cells and using in situ hybridization(ISH)to detect the expression of miR-21-5p and miR-150-5p in Extranodal NK/T cell lymphoma.Combining with relevant clinical symptoms,the prognosis to explore its role in Extranodal NK/T cell lymphoma,and to find new targets related to its therapeutic effect and to provide effective treatment for clinical patients.Methods1 Expression of miR-21-5p and miR-150-5p in NK/T cell lymphoma cell linesThe miRNA specific primers were designed and synthesized by Sangon Biotech(Shanghai)Co.,Ltd.Take U6 specific primers as internal reference,the expression of miR-21-5p and miR-150-5p in human normal NK cells and NK/T cell lymphoma cell lines NK92 and YT were detected by qRT-PCR.2 Expression and clinical significance of mi R-21-5p and miR-150-5p in paraffin tissue of Extranodal NK/T cell lymphoma.2.1 The collection and screening of Extranodal NK/T cell lymphoma of paraffin tissue samples were performed in 40 cases,with 20 cases of chronic inflammation of nasal mucosa as the control group.2.2 The relative expression of miR-21-5p and miR-150-5p in the Extranodal NK/T cell lymphoma and the control tissue of nasal mucosal chronic inflammation was detected by in situ hybridization.2.3 Using SPSS 21.0 software analyze if there are some relationship between the expression of miR-21-5p and mi R-150-5p in Extranodal NK/T cell lymphoma with clinical pathological parameters in patients,including age,gender,B symptoms,serum LDH level,Ann Arbor clinical staging and IPI score.2.4 Single factor and multifactor analysis of the correlation between miR-21-5p and miR-150-5p expression with the above clinical pathological parameters and the clinical prognosis of patients.Results1 The expression of miR-21-5p and miR-150-5p in NK92 and YT of NK/T cell lymphoma cell lines was significantly increased compared with that in normal human NK cell lines.2 Compared with the nasal mucosal chronic inflammation,the expression level of miR-21-5p and miR-150-5p in NK/T lymphoma tissues was significantly higher than that in the control group.3 The expression level of miR-21-5p was not related to age,sex,serum LDH level,B symptoms,Ann Arbor clinical stage and IPI score(P >0.05).The expression level of miR-150-5p was related to IPI score(P < 0.05),but not related to age,sex,serum LDH level and B symptoms and Ann Arbo clinical stage(P > 0.05).4 The expression of miR-21-5p and miR-150-5p and overall survival in patients with no significant correlation(P>0.05),IPI score more than 2 points may be independent risk factors of poor prognosis.Conclusion1 The expression of miR-21-5p and miR-150-5p in Extranodal NK/T cell lymphoma paraffin tissue and NK/T cell lymphoma cell lines was increasing,It is suggested that two kinds of miRNA may be involved in the occurrence and development of Extranodal NK/T cell lymphoma.2 IPI score(? 2 points)is an independent risk factor for patients with poor prognosis,and the level of miR-150-5p expression has a significant correlation with the patient's IPI score.It suggests that the miRNA may be a new prognostic marker for Extranodal NK/T cell lymphoma,and this conclusion needs to further expand the sample size for the prognosis study.
