Experimental Study Of ET-1 Promotes MMP1 And 13 In Chondrocyte Model Of Rat Osteoarthritis

Purpose:To investigate the mechanism of endothelin-1(ET-1)promoting MMP protein up-regulation during osteoarthritis and the correlation of various protein expressions in the conduction pathway.Method:1.Aseptically dissect the hip joint of the mice;scrape the hip joints in a clean bench to show cartilage,and culture the chondrocytes in a cell culture incubator.After successful culture,use an ETRA blocker in a sterile clean bench.(BQ123)and ETRb blocker(BQ788)were used to treat chondrocytes.After half an hour,chondrocytes were treated with ET-1 reagent,and cartilage cells were scraped and frozen to prepare for Western blot.2.After treatment with drugs,each group was divided into:the first group,the normal group(the group without drug treatment);the second group,the ET-1 group of the ET-1 reagent;the third group,the ET-1 and the ETRA Blocking agent(BQ123)reagent BQ123 group;the fourth group,adding ET-1 and ETRB blocking agent(BQ788)reagent BQ788 group;fifth group,adding ET-1,ETRA blocking agent(BQ123)reagent and Group BQ123+788 of the ETRB blocker(BQ788)reagent.3.The expression of the protein of IHH,RUNX2,MMP1 and MMP 13 in chondrocytes treated with ET-1 and its blocker was detected by Western blot..Results:The results of Western blot showed that the expression of IHH,RUNX2,MMP 1,and 1 protein was significantly higher in chondrocytes treated with ET-1 than that in the normal group.The statistical result of each group was P<0.005,which was statistically significant.Compared with ET-1 group,the expression of IHH,RUNX2,MMP1,and 13 protein in chondrocytes of BQ123 group,BQ788 group,and BQ123+788 group was significantly decreased,and the statistical result of each group was P<0.005 with statistical significance.Conclusion:In ET-1 promoted osteoarthritis,hypertrophy of chondrocytes may be caused by IHH and RUNX2 proteins,followed by stimulation of matrix metalloproteinases(MMPs),leading to osteoarthritis.
