At present such traditional drugs as estrogen, calcitoninl, fluorides, calcium and vitamin D, etc are mainly used to treat osteoporosis. Although they have certain curative effect, the side effects of them can't be avoided. Since the traditional Chinese medicine for treating osteoporosis have advantage of significant effects, low cost and minor side effects, they have been gradually applied to the clinic for the treatment of osteoporosis. Among the medicine Epimedium from Berberidaceae Epimedium is one of the common drugs for treating osteoporosis. In this study SD female rats were used, bilateral ovariectomy was made and osteoporosis model was constructed. The experimental rats were fed with icariin, and urine, blood, femoral and lumbar spine were taken. Then DPD, ALP, bone density, bone biomechanics, bone histology and the change of OPG gene were determined. Accordingly the intervention of icariin on bone metabolism in ovariectomized rats was discussed, which provided theoretical and experimental basis for the treatment of post-menopause osteoporosis by using icariin. Three experiments were completed in this study.Experiment 1:Construction and identification of osteoporosis model of rats. 24 female adult SD rats were divided into group A, sham operation group (Sham group), group B, ovariectomized group (OVX group) and group C estrogen group (bilateral OVX + estrogen). The results showed that:ALP value in OVX group increased significantly compared with that in SHAM group; BMD value of lumbar spine and femur in OVX group decreased significantly compared with that in SHAM group. It could be found that cortical bone got thinner, marrow cavity expansed, distribution of trabecular bone was raritas, trabecular became thinner and thickness varied, there were some breakages, and uterine weight of rats in OVX group was 75.5% ligher than that in SHAM group.Experiment 2:Effects of icariin on osteoporosis in ovariectomized rats. Adult female SD rats were divided into:group A, sham operation group (SHAM group), group B, ovariectomized (OVX group), group C, estrogen group (bilateral ovariectomy+estrogen), group D1, low dose of cariin group (OVX+ low dose icariin), group D2, medium dose icariin (OVX+medium dose icariin), group D3, high dose icariin group (OVX+high dose icariin). The experimental results showed that medium and high dose icariin could increase bone mineral density and bone strength of OVX rats. Experiment 3 Effects of icariin on bone marrow gene expression of OPG mRNA in ovariectomized rats:. Adult female SD rats were divided into:group A, sham operation group (SHAM group), group B, ovariectomized group (OVX group), group C, estrogen group (OVX+estrogen), group D1, low dose icariin group (OVX+low dose icariin), group D2, medium dose icariin group (bilateral OVX+medium dose icariin) and group D3, high dose icariin (OVX+high dose icariin). The results showed:The level of OPG mRNA gene expression in OVX group decreased significantly compared with that in SHAM group by semi-quantitative RT-PCR analysis, which indicated that high dose icariin could inhibit absorption of bone minerals strongly.Conclusions:1.3-month-old female SD rats were chosen, the method of ovariectomy were used and animal models of osteoporosis were constructd. It showed serum ALP levels of ovariectomized rats increased significantly compared with that in sham group, bone mineral density of femoral and lumbar decreased significantly compared with that in control group. It also showed that trabecular bone got thinner, the number of cortical reduced and uterine weight was ovbiously lighter than that in control group. The experimental results showed that:the experiment could produce an ideal animal model of osteoporosis and provide an ideal animal model for the use of anti-osteoporosis medicines. Meanwhile, it concluded that estrogen deficiency correlated with postmenopausal definitely..2 Animal models of osteoporosis were interfered by using icariin. The results showed:icariin could significantly increase bone mineral density, bone strength, OPG mRNA expression of ovariectomized rats, reduce concentration of urinary DPD excretion and increase trabecular bone density. It was also thought that the drug icariin had estrogenic effect and could inhibit the loss of bone mass after menopause.3. By using Dual Energy X-ray BMD Apparatus, AUTOGRADH testing machine, semi-quantitative genetic testing and urine DPD testing, bone metabolism could be detected accurately, which was of great value in the early diagnosis of osteoporosis.4 The traditional Chinese medicine has great potential and value in the prevention of osteoporosis. This study proved icariin had good effect in treating rats osteoporosis induced by ovariotomy and revealed icariin played a role in prevention of osteoporosis.
