Objective: 1.To study the effect of modified qifang granule containing serum on AGS cell proliferation in gastric cancer.2.To investigate the miRNA expression and preliminary effect analysis of modified qifang stomachache granule containing serum after interfering with AGS cells of gastric cancer.Methods: 1.AGS cells of gastric cancer were cultured in vitro.2.Rats were fed with modified qifang stomachache granule for 5 days to extract serum containing drugs and serum from healthy rats.3.Blank control group,negative control group(animal serum group)and drug serum intervention group were set up.Negative control group with animal serum;The drug serum intervention group was divided into 5 concentration gradients,and the final concentration was 2.5%,5%,10%,15% and 20% respectively.Culture in the incubator for 24 h,48h and 72 h respectively.The effects of different concentrations of serum containing drugs on the proliferation of gastric cancer cells were detected by cck8.4.Blank group: AGS cells of gastric cancer were routinely cultured.Negative control group: AGS cells of gastric cancer were added to normal rat serum of the same dose as the drug observation group for cell culture.Experimental group: AGS gastric cancer cells,flavored seven square stomach granule drug-containing serum intervention,intervention after 72 hours,the extraction of total RNA and quality control,and then build a library,high-throughput sequencing,the expression of micrornas difference spectrum analysis.Results: 1.The seven parties flavored stomach granule drug-containing serum intervention AGS cells after 24 h,48 h,72 h,the concentration group of AGS have inhibition of proliferation,compared with negative control group differences statistically significant(P < 0.05),and the time and the concentration dependence.2.After AGS cells of gastric cancer were treated with modified qifang stomachache granule,22 mirnas were up-regulated and 23 mirnas were down-regulated.The 22 upregulated mirnas were: Mir-14-5p,mir-345-5p,mir-509-3p,let-7c-5p,mir-411-5p,mir-99a-5p,Mir-1286,mir-135a-5p,mir-3620-3p,mir-1-3p,mir-513a-5p,mir-378 f,mir-3138,mir-3180,mir-3180-3p,mir-379-5p,mir-654-5p,mir-6747-3144-3p,mir-468-3p;23 down-regulated mirnas were: Mir-1261,mir-497-5p,mir-512-3p,mir-525-5p,mir-548az-544 p,mir-67355-5p,mir-675-3p,mir-219b-3p,mir-2277-3p,mir-5180-5p,mir-5180-5p,mir-12526-43-1.83.Mir-4802-3p,mir-510-5p,mir-3158-5p,mir-7114-5p.Conclusion: 1.The serum containing qifang stomachache granule has an inhibitory effect on the proliferation of AGS cells in gastric cancer,which is dependent on time and concentration.2.After the intervention of modified qifang stomachache granule in AGS cells of gastric cancer,differential expression of miRNA appeared,which may Inhibit the proliferation of gastric cancer cells by causing differential expression of mir-144-3p,mir-99a-5p,etc.
