| Objective This study through identifying the miRNA which is closely related tohuman gastric cancer, to probe the possible pathogeny and preliminary making up theexpression profiling for miRNA of different differentiation degree of human gastriccancer, to lay the groundwork for further research on the mechanism of the emergenceand development of gastric cancer and biomarker which is related to the diagnosis andtreatment of gastric cancer.Methods1. Culture human gastric cancer cell line NCI-N87, MGC-803and normalgastric epithelium cell GES-1in vitro, extract the total RNA of these cells, differentiallyexpressed miRNA were filtered using miRNA microarray technoloygy. Some ofdifferentially expressed miRNA were verified by qRT-PCR.2. Selecting the specificmiRNA, and to validate their expression in27cases of gastric cancer and matchednon-tumor tissue.Results1. miRNA microarray detection that there were147miRNAs with twice andmore different expression in NCI-N87and MGC-803by compared with GES-1. Andthere were54miRNAs were significantly up-regulated, and58were significantlydown-regulated in NCI-N87. And there were42miRNAs were significantlyup-regulated, and33were significantly down-regulated in MGC-803. The findings ofqTR-PCR on3miRNAs(miR-16of up-regulation and miR-7, miR-185ofdown-regulation) reveal that most of the microarray results credible.2. It was revealedby miRNA microarray that part of virus-associated miRNA with different expression,such as kshv-miR-K12-12*, ebv-miR-BART19-3p, hsv1-miR-H7and et al.3. Study of27cases of patients with gastric cancer and adjacent non-tumor tissue miR-7expressionlevels found, compared to paired adjacent non-tumor tissue,19cases of patients withgastric cancer, the expression of miR-7down-regulated, statistical analysis showed this difference was statistically significant(t=-2.809,P<0.05), indicating that the miR-7expression in tumor tissue compared with paired non-tumor tissues was reduced.Conclusions1. Initial screening of147miRNAs which related with human gastriccancer, and different differentiation degree of gastric cancer cells are present specificmiRNA expression profiling, to provide the relevant information for building a newmethod of diagnosis and treatment for gastric cancer.2. Virus-associated miRNAdifferent expression in human gastric cancer cell lines and GES-1suggests that part ofthe virus miRNA may be involved in the incidence and development process of gastriccancer.3. miR-7is possible down-regulated in hunman gastric cancer and may as thetumor suppressor gene involved in the emergence and development process of gastriccancer. |
