Objective: To investigate the expression levels of sclerostin(sclerostin,SOST)?TNF-??IL-1??IL-6 in the plasma of arthritis(rheumatoid arthritis,RA)patients;to investigate the correlation between SOST?TNF-??IL-1??IL-6 and the clinical commonly used detection indexes(GLOB,ESR,RF,CCP etc.)of RA patients.Methods:(1)Collecting the clinical datas and laboratory indexes(GLOB,ESR,RF,CCP,etc.)and peripheral venous blood specimens from plasma of 60 RA patients(group RA)(30 men(group RA(M),30 women(group(RA(F))and 60 healthy subjects(group HC)(30 men(group(HC(M),30 women(group HC(F)).(2)Detecting the expression levels of SOST?TNF-??IL-1?and IL-6 in each group of plasma through ELISA(enzyme linked immunosorbent assay).(3)Using the SPSS 17.0 statistical software to analyze,using Wilcoxon test and one-way analysis of variance analysis to compare among measurement groups,using Spearman to analyze the correlation among variables.Results:(1)In plasma,the level of SOST in group RA(1.50±1.24 ng/ml)was higher than that in group HC(1.29±0.77ng/m),no statistical significance(P>0.05);after dividing group RA and group HC into groups according,to the gender,the level of SOST in group RA(F)(1.69±1.16ng/ml)was trendingly higher than that in group HC(F)(1.17±0.77ng/ml),no statistical significance(P>0.05);there was no significant difference between the level of SOST in group RA(M)(1.31±1.30ng/ml)and group HC(M)(1.41 ±0.76ng/ml),no statistical significance(P>0.05).(2)In plasma,the levels of TNF-??IL-1??IL-6 in group RA(11.43±4.28pg/ml?4.51±8.16pg/ml?12.07±19.14pg/ml)were higher than those in group HC(10.12±4.92pg/ml?3.13±1.48pg/ml?1.36±2.67pg/ml),statistical significance(P<0.05);the levels of TNF-?? IL-1? ? IL-6 in group RA(M)(12.78±2.94pg/ml ? 5.04±10.91pg/ml ?15.09±24.67pg/ml)were all significantly higher than those in group HC(F)(11.48±6.48pg/ml?3.31±1.71pg/ml?1.23±1.30 pg/ml),those in group HC(M)(8.77±1.87pg/ml?2.94±1.2pg/ml?1.49±3.57 pg/ml)and those in group RA(F)(10.09±4.99pg/ml?3.98±3.97pg/ml?9.05±10.85pg/ml),statistical significance(P<0.05);the levels of TNF-??IL-1??IL-6 in group RA(F)were significantly higher than those in group HC(F)and group HC(M),comparing TNF-? and IL-1? between group RA(F)and group HC(F)?group HC(M),no statistical significance(P>0.05);comparing IL-6 between group RA(F)and group HC(F)?group HC(M),statistical significance(P<0.05).(3)In the analysis of clinical trial datas,the levels of GLOB?ESR?RF?CCP in group RA(F)(36.0±6.1g/L?33.2±24.0mm/h?172.8±293.3IU/ml?113.1±52.5U/ml)were all significantly higher than those in group HC(F)(31.6±4.8g/L?4.5±2.0mm/h?12.0±5.1IU/ml?3.1±1.6U/ml),the levels of GLOB?ESR?RF?CCP in group RA(M)(34.8±5.0g/L?38.0±32.1mm/h?174.3±150.4IU/ml?95.0±48.7U/ml)were all higher than those in group HC(M)(26.4±3.8g/L?4.0±1.8 mm/h?11.6 ±4.5IU/ml?3.0±1.7U/ml),statistical significance(P<0.05).(4)For RA patients,it was found through Spearman correlation analysis that SOST in group RA(F)was negatively correlated with CCP(r=-0.416,P=0.022),SOST in group RA(M)was positively correlated with ESR(r=0.564,P=0.001),SOST was positively correlated with IL-6(r=0.707,P=0.000),SOST in group HC(M)was positively correlated with IL-1?(r=0.402,P=0.028),In each group,there was no significant correlation between SOST and other factors(P>0.05).Conclusion: The expression levels of SOST?TNF-??IL-1?and IL-6 in the plasma of RA patients are higher than healthy subjects.In addition,the expression level of SOST was correlated with its inflammatory indexes(ESR?CCP and IL-6),suggesting that SOST may take part in the happening of RA and makes a difference to its inflammatory reaction.
