Background and ObjectiveOvarian cancer is a common malignant tumor in gynecology,Of these,more and more cutaneous carcinomas are common,The mortality rate is high in the female reproductive system malignant tumor.In recent years,with the continuous improvement of operative technology and new chemotherapeutic agents,the therapeutic effect of epithelial ovarian cancer has been improved significantly,but the5 year survival rate has not yet been significantly improved.Therefore,searching for tumor markers related to the development and prognosis of epithelial ovarian cancer and effective intervention is the urgent problem at home and abroad.SND1?staphylococcal nulease domain containing 1?,It is also known as P100,or Tudor-SN?tudor staphylococcal nulease?,It was discovered for the first time that the transcriptional regulation activator of the EB virus nuclear antigen 2 was found by Tong and so on.Widespread in a variety of organisms,such as human beings,animals,plants,etc.Highly conserved among different species.It is found that SND1 can be involved in many biological processes such as cell signal transduction,RNA splicing,cell proliferation,apoptosis,cell stress,viral infection,tumor occurrence and so on.The high expression of SND1 is involved in the development of many kinds of tumor,such as breast cancer,liver cancer,prostate cancer and so on.However,whether the expression of SND1 in ovarian cancer is abnormal and its abnormal expression is related to the development of ovarian cancer,now few studies have been made at home and abroad.In this article,the immunohistochemical staining method and Western imprinting method was employed to assay the expression of SND1 protein in the specimens of50 cases with epithelial ovarian carcinoma,20 cases with borderline ovarian cystadenomas,and 20 cases of normal ovarian tissue.Reverse transcription polymerase chain reaction was used to determin SND1mRNA expression.And the relationship between the expression of SND1 and the pathological stages,histological differentiation,lymph node metastasis and other clinical features of ovarian cancer were studied.To explore the role of SND1 in the development of epithelial ovarian cancer.Materials and Methods1.Material source:Collect epithelial ovarian cancer from 2016 September to2017 October in the First Affiliated Hospital of Zhengzhou University underwent cytoreductive surgery for ovarian cancer or comprehensive surgical staging of primary 50 patients,including 18 cases of mucinous cystadenocarcinoma,26 cases of serous cystadenocarcinoma,6 cases of endometrial adenocarcinoma.The age was44-68 years,and the average age was?56±12?years.No preoperative radiotherapy,chemotherapy,hormone and immune therapy;the other surgical resection of borderline ovarian tumor in 20 cases,due to other reasons for oophorectomy 20 cases of normal ovarian tissue as the control group.About 2 pieces of active tissue at the edge of the tumor tissue were collected from each group,about 1cm3 size.2.Experimental method:The immunohistochemical staining method and Western imprinting method was employed to assay the expression of SND1 protein in the specimens of 50 cases with epithelial ovarian carcinoma,20 cases with borderline ovarian cystadenomas,and 20 cases of exact probability method ofFisher,?2test and continuous proofreading of?2test,the diffirence of the positive rate of the expression of SND1protein in different groups was tested.The correlation date of the expression of SND1protein was analyzed with 2×2 matching data.The test level a=0.05,P?0.05represents the difference statistical sigificance.Results1.The positive reaction of SND1 protein immunohistochemical staining was brown yellow granules,which was mainly located in the cytoplasm.SND1 protein in epithelial ovarian cancer tissues the positive rate was 88%,significantly higher than the positive expression rate in ovarian borderline epithelial tumor tissues?55%,?2=8.947,P<0.05?,which was significantly higher than that in normal ovarian tissues.The positive expression rate?25%,?2=27,P<0.05?,the difference was statistically significant;while the positive expression rate of SND1 in ovarian borderline epithelial tumor tissues than in normal ovarian tissues the expression of positive rates between the groups??2=3.75,P>0.05?,the difference was not statistically significant.2.The high expression of SND1 mRNA in ovarian cancer tissues was significantly higher than that in borderline ovarian tumor and normal ovarian tissues?P<0.05?,the expression of borderline ovarian epithelial tumors and normal ovarian tissues showed no significant difference?P>0.05?.3.The high expression of SND1 protein in ovarian cancer tissues was significantly higher than that in borderline ovarian tumor and normal ovarian tissues?P<0.05?,the expression of borderline ovarian epithelial tumors and normal ovarian tissues showed no significant difference?P>0.05?.4.According to the age,pathological stage,histological differentiation and lymph node metastasis score showed that SND1 protein in ovarian epithelial cancer?III+IV?is higher than the expression?I+II???2=7.882,P<0.05?;in poorly differentiated epithelial ovarian carcinoma expression was higher than that in the high differentiation and differentiation of epithelial ovarian cancer??2=7.071,P<0.05?;SND1 protein expression in patients with lymph node metastasis than in patients without lymph node metastasis in lymph increased significantly??2=8.612,P<0.05?.Conclusion1.The expression of SND1 protein and SND1 mRNA in epithelial ovarian cancer tissues is high,and SND1 may play a role in the development of epithelial ovarian cancer.2.The expression of SND1 protein is closely related to the pathologic stages of epithelial ovarian cancer,the degree of tissue differentiation and lymph node metastasis.
