The content of flavonoids in onion bulbs is reduced from outside to inside in turn,peel is the highest level.In order to take full advantage of the flavonoid-riched onion peel,reduce the waste of resources,onion peel was studied as raw material.Flavonoids were extracted from onion peel by solvent extraction method in this experiment,then concentrated and lyophilized into powder,eventually applied into the research of anti-obesity.4?5 week-old SPF C57BL/6J mice were tested in this study,and randomly divided into 6 groups after 1 week adaptive feeding.The breeding cycle of 10 mice each group was 10 weeks.The normal control group was given distilled water by lavage once a day with ordinary diet simultaneously,the other groups were fed high-fat diet with different lavage.The experimental groups were given by lavage through high,medium and low dose(6.6%,3.3%,1.67%)orally of onion peel extract,and the positive control group was given 3.3%polyphenols.The main experimental results were as follows:a)The process of weight gain in mice was slow to a certain extent by both onion peel extract and polyphenols,the body fat accumulation in mice were also reduced.Surprised to find that,the effect of inhibiting weight gain by a dose of onion peel extract was significantly better than polyphenols.Lee index and coefficient fat of experimental groups and the positive control group were lower than the high-diet control group,there was a significant difference(p<0.05);Liver index of the high-diet control group was significantly higher than the normal control group(p<0.05),liver index of the experimental groups and the positive control group was significantly lower than the high-diet control group(p<0.05),decreased by 26.8%and 25.1%separately,what indicating that the deposition of fat in the liver were inhibited by onion peel extract obviously.The cardiac index,spleen index,and kidney index between each test groups were not significantly different(p>0.05).b)The level of TC,TG and LDL-C in mice serum of the experimental groups were significantly lower than the high-diet group(p<0.05),and the content of HDL-C was higher than high-diet group,a significant difference was shown(p<0.05);The level of T-AOC was improved significantly by medium and high dose onion peel extract,the antioxidant enzymes activity of SOD and GSH-Px were increased significantly,the lipid peroxide content of MDA decreased,and a significant difference was shown compared with high-diet control group.In summary,the lipid balance was regulated and the body's antioxidant effect was enhanced by onion peel extract.c)Compared with the normal control group,the morphology of adipocyte of the high-diet control group were significantly inflated,the number of adipocyte in the same field of microscope vision was reduced.The adipocyte of positive control group and the low-dose group were in different sizes,however,the size of adipocyte were reduced to a certain extent.The adipocyte morphology of high-dose group and mid-dose group were in a similar size,and the diameter of adipocyte were significantly smaller than high-diet control group,the number of adipocyte in the same field of view was increased,what's more,the results were better than the positive control group.It shown that the accumulation of adipocyte in fat could be reduced by onion peel extract,and the adipocyte enlargement could be inhibited also.d)The morphology and size of hepatocyte in high-dose group and mid-dose group were normal,hepatic cord neatly arranged ordered,and the structure of hepatic lobule was normal,no significant difference compared with the normal control group.Inflammatory cell infiltration of hepatocyte were visible in high-diet control group,vesicle fatty infiltration were increased,what can be considered as liver steatosis.Visibly,the fat accumulation amount in mice liver of experimental groups were better than high-diet control group.
