Viscum col or at um(Kom.)Nakai f.Rubroaurantiacum Kitag(VCR)is a kind of common traditional herbal medicine in China,which belongs to genus Viscum coloratum of the Loranthaceae family.It is distributed widely across our country,especially in Northern China.The medicinal part of Viscum is dry stem with leaves.The plants of genus Viscum show several kinds of medicinal effects,such as rheumatic paralysis,metrorrhagia,blood leakage,dizziness,fetal irritability,nourishing blood for preventing miscarriage,etc.Modern research found that it has the effects of antibacterial,anti-ageing and immunomodulatory,in addition to have.a variety of different levels of tumor inhibition.Vi scum coloratum not only have the biological active components of large molecules,such as polysaccharides,lectins,also contains the small molecules,such as alkaloids constituent.The experiments demonstrate that,it has in vivo tumor destruction.Viscum coloratum is a mixture of a variety of alkaloids,which has antitumor,low toxicity and low cost characteristics.Therefore,the study on the extraction,separation and purification of alkaloid from mistletoe,it can make full use of the rich resources in the north part of country,but also provide the theoretical guidance for further development.The pharmacokinetic analysis to Viscum coloratum,which to discribe the concentration of drug in the body along with the time variation of the dynamic process,it can guide the development of new drugs and it has has very important significance to clinical rational drug.In this study,mistletoe alkaloid extraction process is preferably used ion exchange resin,By orthogonal experiment to determine the best extraction process.the most optimum:acid infiltration percolation concentration was 2 per thousand,infiltration percolation liquid dosage is 15 times of the medicinal material,resin dosage is 0.5 times of the herbs ammonia amount of per 15mL/100g.It has a high efficiency extraction and get a better validation in the test.Using acid dye than The average content of total alkaloids of color method for0.379mg/g.By silica gel column chromatography to separate and purify mistletoe alkaloid,N-butyl amido cinnamyl amine and N-cinnamoyl spermidine is obtained after separation and purification,which purity above 98%by the characterization of MS,1H-NMR,13C-NMR and HPLC.And the pharmacokinetic studies on the N-butyl amido cnnamyl amine,and N-cinnamoyl spermidine,it can establ ish a simple,sensitive and specific blood drug concentration by HPLC determination method.Through to gavage the three different doses,the blood drug concentration time curve showed a certain degree of similarity.The pharmacokinetic characteristics of one compartment model.After administrate three different doses(25mg·kg-1,50m g·kg-1,75mg·kg-1)of N-butyl ami do cinnamyl amine,the reaching peak time was 1.000?0.750?0.750h,clearance(CL)was 5.840,11.172,12.741L·h-1·kg-1,the apparent volume of distribution was 13.884,21.047,35.697L·kg-1;After administrate three different doses(25 mg·kg-1?15 0mg·kg-1?75mg·kg-1)of N-cinnamoyl spermidine,the reaching peak time was 0.750h,clearance(CL)was 15.828,16.470,19.069L·kg-1,the apparent volume of distribution was 9.360,14.171,15.687 L·h-1 · kg-1.After administrate the intestinal absorption and clearance in the plasma is very quike,and the distribution is broadly.
