| Purpose:Radix Aconiti kusnezoffii is the dried root of Aconitum kusnezoffii Maxim, which is Ranunculaceae Aconitum plants. The main components of its efficacy are Aconitum alkaloids, with Qufeng dehumidification, warming the channels to relieve pain and other effects, but it has great toxicity. Research has shown that, the toxicity of alkaloids is larger for the diester alkaloids, prone to hydrolysis, the hydrolysis products of the monoester Aconitum analgesic anti-inflammatory effect is still good, toxicity is greatly reduced. This paper aims at extracting method to establish a rapid, simple, and efficient method for the analysis of alkaloids, through the qualitative analysis and the fingerprint, to study the chemical constituents in the hydrolysate, lay the foundation of quality evaluation method and low toxicity and efficacy components for Aconitum medicinal materials.Method:Diester and monoester Aconitum alkaloids extract, hydrolyzed by high performance liquid chromatography and spectroscopy combined with technical means of identification, deepen its chemical components, to establish the standard fingerprint, analysis of different batches of Radix Linderae material grass by the similarity and multivariate statistics, and these three kinds of single ester type alkaloids in rats with adjuvant arthritis treatment.Result:Separation using chromatography, yielded four compounds from the extract of Radix Aconiti kusnezoffii. Respectively, they are aconitine, mesaconitine, hypaconitine, and deoxidation. The four compounds after further hydrolysis, eleven compounds were obtained. They are benzoylaconine, aconine, benzoylhypaconine, mesaconine,benzoylmesaconine, mesaconine,benzoyldeoxyaconine,hypaconine,pyroaconitine,pyromesaconitine,pyrohypaconit ine, pyrodeoxyaconitine. On the basis of fingerprint and Radix Aconiti kusnezoffii are analyzed, evaluated the content difference of the main compounds. Reviewing biological activity of benzoylmesaconine, benzoylaconine, benzoylhypaconine, the results show, they are able to reduce rat primary and secondary paw swelling rate, and the amount of adjuvant arthritis rat ESR and WBC.Conclusion:This paper studied on the chemical constituents of Radix Aconiti kusnezoffii, Provides a rapid and scientific evaluation method for the fingerprint. Finally, by research of aconitine monoester of bio-activity, the preliminary results show that it has certain effect in treatment of rheumatoid arthritis. Therefore, the research on the level of theory and practice, has the vital significance. |
