Animal stress has always been one of the hot spots in the animal husbandry industry.Heat stress is an important factor affecting the production and reproductive performance of livestock and poultry,also could induce various diseases.Xiangchunzi is the fruit of Toona sinensis(A.Juss.)Roem.Many studies have shown that Toona sinensis fruit contains flavonoids,phenolic acids,terpenes,alkaloids and other active ingredients.It is a natural antioxidant and has many functions,such as treatment of atherosclerosis,antibacterial and anti-inflammatory,lowering blood sugar,protecting liver,anti-gout,relieving liver cirrhosis,improving memory.In this study,Toona sinensis fruit were used as raw materials to obtain extracts from different parts of Toona sinensis fruit(husk,fruit shaft and wing membrane)using water extraction and ethanol extraction methods.The qualitative,quantitative and in vitro antioxidant capacity tests were carried out.Then the papper uesed Caenorhabditis elegans as a model organism,heat stress at 35? to explore the influence of different parts of Toona sinensis on the life of Caenorhabditis elegans.Finally,the alcohol extract of Toona sinensis husk was used to treat the nematodes on heat stress,and the expression of daf-2,sod-3,jnk-1 ? sir-2.1 were detected to explore the possible mechanism.The research results are as follows:1.Qualitative,quantitative and antioxidant research on the active substances in different parts of Toona sinensisThe husk,fruit shaft and wing membrane were separated from the Toona sinensis fruit,dried and crushed to obtain the extract using water extraction and ethanol extraction methods.The qualitative,quantitative and in vitro antioxidant capacity were tested,and different parts of the Toona sinensis fruit were found.The content ofphenolic acids in the extracts were significantly different.The alcohol extract of fruit husk contained the most phenolic acids.The antioxidant capacity of Toona sinensis wing membrane water extract and fruit husk alcohol extract were higher than that of other parts.2.The effect of water extract of Toona sinensis wing membrane and alcohol extract of fruit husk on the life span of heat-stressed nematodesBoth the water extract of Toona sinensis wing membrane and the alcohol extract of husk could significantly increase the survival rate of nematodes treated with heat stress at 35?,and the alcohol extract of fruit husk had a better protective effect on nematode heat stress(P <0.01).3.The effect of alcohol extract of Toona sinensis husk on the movement ability of nematodes under heat stressA certain concentration of alcohol extract from the husk of Toona sinensis can enhance the head oscillation and body bending frequency of nematodes.4.The effect of Toona sinensis husk extract on the expression of heat-stressed nematode related genesAfter treating the nematodes at 35?,compared with the control group,the genes expression of the nematodes in the Toona sinensis husk treatment group was changed,the jnk-1 gene was up-regulated and the difference was significant(P <0.05);the sod-3 gene was up-regulated and the difference was extremely significant(P <0.01);the daf-2 gene and sir-2.1 gene were down-regulated and the difference was extremely significant(P <0.01).
