The density of Muscovy Duck semen is high.Before artificial fertilization,it is often necessary to dilute the collected semen in a certain proportion to improve the semen utilization rate.In this study,the semen of Muscovy duck was diluted with different diluents at different dilution ratios and kept at room temperature and low temperature respectively.At the same time,artificial insemination was carried out after diluting at 1:1 ratio.The best diluent and dilution ratio for normal and low temperature preservation of Muscovy Duck semen and the best diluent for artificial insemination were selected.This study consists of four parts:The purpose of this test is to select the best dilution,dilution ratio and storage temperature suitable for storing muscovy duck semen at normal temperature(15-25℃).The test was divided into four dilutions of NO-1,NO-2,BJJX and saline,respectively,in the ratio of semen-diluent v/v: 2:1,1:1,1:2,and respectively at 15℃,17℃,20℃,25℃ constant temperature storage.The sperm motility,sperm deformity and plasma membrane integrity were measured every 8 hours from 0 h.The test results show that the NO-1 buffer the best diluent for storage at room temperature;the storage time and storage quality of semen at 25℃ is significantly lower than 15~20℃;compared with the 2:1 ratio,the better quality duration was observed in samples diluted at the 1:1 ratioThe purpose of this test is to determine the appropriate dilution ratio and dilution of Muscovy duck semen stored at 4℃.In the experiment,four dilutions of NO-1,NO-2,BJJX and saline were respectively diluted with semen-diluent volume ratio of 2:1,1:1,1:2,and kept in a refrigerator at 4℃ constant temperature.The sperm quality was measured every 8 hours.The test results showed that NO-1 showed the best storage quality and the longest storage time at a 1:1 ratio(72±8.00 h).The purpose of this test is to use NO-1 as the base fluid and add different amounts of skimmed milk powder to determine the optimal content of skimmed milk powder in Muscovy duck semen stored at 4 ℃.In the experiment,5 concentrations of skimmed milk powder(0.02,0.04,0.06,0.08,0.10 g)were added respectively,and the sperm motility,plasma membrane integrity rate and sperm deformity rate were measured every 8 hours,and the storage time was measured.The results show that 0.08 g of skimmed milk powder can significantly prolong the sperm storage time(117.33±4.62),significantly reduce the proportion of deformed sperm,improve the sperm survival rate and plasma membrane integrity rate,showing the best preservation effect.In this experiment,four dilutions of NO-1,NO-2,BJJX and saline were diluted at a 1:1 ratio of semen-diluent,and 1080 black black female muscovy ducks were artificially inseminated.The experiment shows that the experience groups can significantly increase the fertilization rate,compared with saline group(65.35±1.01),and the NO-1 group has the highest fertilization rate(70.25±0.22),which is the best dilution for artificial insemination of muscovy duck semen.In summary,the quality of semen stored at low temperature is significantly affected by the composition of the diluent,the dilution ratio and the storage time;the addition of skim milk powder in the diluent can significantly prolong the storage time of the semen,and the optimal amount is 0.08 g. |
