In recent years,the global slow a high incidence of disease and epidemic diseases is increasing,more and more people have insomnia phenomenon,insomnia has become epidemic in the world today,according to relevant data statistics,45% in China,Europe and the United States there is a 10%-more than 30% of people sleep disorder,insomnia and rise by a spurt in recent years,the serious influence health,work and property safety.Actinidia argute Sieb.et Zucc is rich in various nutrients and vitamin C.its special secondary components have good curative effect on most human diseases?such as diabetes,hypertension,hyperlipidemia,etc.?.1.Actinidia argute Sieb.et Zucc has different characteristics due to different production areas and strains.The fruit shape of the Actinidia argute used in this experiment is mainly oblong and oblong,with a single fruit weight ranging from 5.82 to 13.81 g.Among them,the range of soluble solids is between 11.70% and 17.30%,the content of vitamin C is between 352.61 and 563.61 mg·g-1,the content of titratable acid is between 0.12% and 0.30%,and the content of reducing sugars is between Between 3.42% and 7.62%.These ten strains of soft date kiwi all contain terpenes,phenols,tannins,volatile oils,lipids,flavones and their glycosides,polysaccharides and their glycosides,and they do not contain saponin.2.Analysis of the results by infrared spectroscopy revealed that the polysaccharides of the 10 strains all contained-NH2,-OH or-NH-groups.The differences between the different strains are as follows: strains 3,4,6,7,9 are dominated by ?-type glycosidic bonds;strain 5 is dominated by ?-type glycosidic bonds;strains 5,6,7 are pyranosidic bond types;Strains 10 are all acidic polysaccharides;strains 5,6,7,10 have more polysaccharides;strains 1,4,and 8 have lower polysaccharides;the other strains have no significant differences in polysaccharide content;strains 1,6,7,8,and The peak shape of 9 is a convex peak,and the peak shapes of lines 2,3,4,5,and 10 are stepped peaks.Polysaccharide functional groups and convex peaks containing-NH2,-OH,-NH-are involved in the regulation of insomnia in mice.3.Extraction,separation and detection of flavonoids in Actinidia kiwifruit.Based on the analysis of the spectrum,12 compounds were identified through preliminary comparison of excimer ion peaks and multi-level mass spectrometry fragment information with standards.They are rutin,quercetin,rhamnoside,chrysanthesin,diglycoside,xanthoside,pyran-glucoside,kaempferol-glucoside,gallate,flavanone,daumone,?-Balsamic resin alcohol acetate and other substances.Rutin,daimone,and rhamnoside flavonoids contained in all 10 strains are involved in the regulation of insomnia in mice.4.In the experiment of determining the dosage of flavonoids from kiwifruit polysaccharides,it was determined that the compound flavonoids from kiwifruit polysaccharides with high doses(200 mg · kg-1)had the best effect in treating insomnia in mice.That is,the contents of 5-HT and 5-HIAA were significantly increased?P <0.01?,the effect of promoting sleep was significantly reduced,the contents of DA and NE were significantly reduced?P <0.01?,and the effect of inhibiting insomnia was obvious.5-HT is a neurotransmitter that can promote sleep.5-HIAA is an acidic metabolite of 5-HT.The increase in the content of 5-HT and 5-HIAA indirectly indicates that the polysaccharides and flavonoids of jujube kiwifruit can promote sleep.DA has the effect of maintaining arousal and behavioral excitement,NE has an antagonistic effect on 5-HT to maintain sleep,and the reduction of DA and NE content indirectly indicates that the soft date kiwi polysaccharides and flavones can inhibit insomnia,that is,treat insomnia in mice Role.5.In the Actinidia argute polysaccharides and flavonoids treatment insomnia comparison test,the 5-HT content of the flavonoid group 7 increased significantly?P <0.01?,and the 5-HIAA content of the flavonoid group 1 increased significantly?P <0.01?.)That is,the effect of promoting sleep is obvious;the DA content of the polysaccharide group No.1 is significantly reduced?P <0.01?,and the NE content of the flavonoid group No.1 and the flavonoid group No.7 is significantly reduced?P <0.01?,that is,the effect of inhibiting insomnia is obvious.6.Through analysis of different combinations of polysaccharides and flavones,and between different strains,it is concluded that the effect of preventing and treating insomnia in mice is increased by increasing the content of 5-HT and 5-HIAA and decreasing the content of DA and NE.The flavonoid group of line 1 had the most significant effect.
