Sanguisorba Radix is dried roots for Sanguisorba officinalis L.or S.officinalis L.var.longifolia?Bert.?Yüet Li;the taste is bitter,sour and puckery;slightly cold;distributes to liver and large intestine.It has the effects of cooling blood and detoxify,hemostasis and converge sores.It is mainly used to treat blood flux,epistaxis,uterine bleeding,sore and furunculosis.Modern research showed that Sanguisorba officinalis mainly contains tannins,phenolic acids,saponins,flavonoids and polysaccharides.It has antidiarrheal,antibacterial,and anti-inflammatory effects.The present study established the method for detecting gallic acid content in Sanguisorba officinalis extract,optimized the preparation process of Sanguisorba officinalis extract,and investigated the antibacterial effect of Sanguisorba officinalis extract.The study was aimed at providing scientific basis for further formulation development and clinical application.?1?Establishment of method for detecting gallic acid content in Sanguisorba officinalis extract:The high-performance liquid chromatography?HPLC?method was used to study the method for detecting gallic acid in the extract of Sanguisorba officinalis and Sanguisorba officinalis.The results showed that Shim-pack GIST C18?250mm×4.6mm,5?m?was chromatographic column;methanol-0.05%phosphoric acid solution?5:95?was mobile phase;detection wavelength was 272nm;the flow rate was 1.0m L/min;the column temperature was 25?;and the injection volume was 20?L.And the characteristic peak resolution of gallic acid was 2.0,and the theoretical plate number was 3500.In the concentration range from 7.264 to 72.640?g/m L gallic acid showed a good linear relationship with the peak area value.The linear equation was y=62174.2214x-28549.9344?r=0.9999?,the average recovery rate was 98.40%,and RSD=1.06%.This method can be used to determine the content of Sanguisorba officinalis extract.?2?Optimize preparing process of Sanguisorba officinalis extract:On the basis of single-factor test,the response surface method was used to optimize the material-liquid ratio,extraction number and extraction temperature.The results showed that the best preparation process for Sanguisorba officinalis extract was:ethanol concentration 40%,material-liquid ratio 1:30,extraction temperature 90?,extraction twice,once for 1h.Test results showed that under this condition,the dried extract yield,gallic acid transfer rate and the average value of the comprehensive score were 50.76%,118.11%and 97.91%,respectively.These were almost consistent with the predicted value.Above results demonstrated that this condition is the best technological condition for preparing Sanguisorba officinalis extract.?3?Antibacterial effect of Sanguisorba officinalis extract:The antibacterial effect of Sanguisorba officinalis extract on Staphylococcus aureus,Escherichia coli and Salmonella was determined by agar plate punching method.The micro broth dilution method and the scribe method were used to determine the MICs and MBCs.The results showed that the average inhibition zone diameters of Sanguisorba officinalis extract against Staphylococcus aureus,Escherichia coli and Salmonella standard strains were 16.0,17.9and16.6mm;and MICs and MBCs were 0.78mg/m L,respectively.The diameter of the inhibition zone for clinical isolated of Staphylococcus aureus,Escherichia coli and Salmonella were 18.3,13.7and 17.0mm,respectively;MICs were 0.78,6.25 and 0.78mg/m L;MBCs were 0.78,25and 0.78mg/m L.The extract of Sanguisorba officinalis showed significant inhibitory effect on Staphylococcus aureus,Escherichia coli and Salmonella.
