Compared with traditional breeding methods,radiation mutation breeding has the advantage of increasing the mutation rate of species and shortening the breeding period.Though gene mutations it can change a series of biological characteristics of species.Soil,water,light,temperature and other factors are closely related to plant growth and development.The main work is to study effect of radiation on the expression of phytochrome B gene in maize.During this process by using the methods of radiation mutagenesis technology,molecular biology experiments and field biological traits detection,the phytochrome gene expression in maize under different light conditions were studied.The relationship between gene expression and plant height were analyzed.The results show:In terms of apparent traits,under the same light,with the increase of dose,the seedling height of each group has variously changed.In terms of gene expression,ionizing radiation and light condition can induce changes in the expression levels of ZmPhyB1 and ZmPhyB2 in maize,and they have different effects on plant height.The seedling of maize at each dose was basically the lowest plant height in the white light environment,lower than the blue light,lower than the red light and darkness.Moreover,the relative expression level of the two genes in white light and dark conditions was significantly higher than that in red light and blue light conditions.The plant height of maize seedlings under white light,red light and blue light had a negative correlation with the expression of ZmPhyB1 in the plant height,and there was a positive correlation in darkness at the dose 10-40 Gy.Another work attended is to study Morphological Changes and RAPD Analysis the M2 Generation of Maize named baimaya radiated by proton.Compared with the control,the germination rates of 20 Gy group and 30 Gy group were higher than those of the control and the highest was 20 Gy group.In terms of the empty rod rate,the lowest were 20 Gy group and 50 Gy group,the highest was 29.4% of the 40 Gy group.The photosynthetic rate of radiation group was significantly lower than that of the control.The 1000-grain weights of 10 Gy group,20 Gy group,and 40 Gy group were significantly higher than the controls and 50 Gy group lower than the control.Considering the germination rate,empty rod rate and 1000-grain weight,the radiation dose with more beneficial variation on M2 generationof baimaya corn was 20 Gy,10Gy and 30 Gy in turn.In RAPD experiment results analyze,the polymorphism rate was 57.3%,the variation rate of each radiation group was between20.2% ~ 32.3%.The result of genetic similarity clustering analysis showed that those groups were clustered into two categories.One included the 10 Gy group and the 50 Gy group,and the variation was high.The other included the 20 Gy,30Gy and 40 Gy groups and the control.
