Ophioordyceps sinensis is a traditional Chinese medicine.Driven by interests,the unsustainable utilization of Ophioordyceps sinensis resources and the destruction of fungus source are caused.Therefore,the artificial fermentation of Ophioordyceps sinensis is becoming an important means to protect the wild resources of Ophioordyceps sinensis.It is a practical requirement to select high quality strains and evaluate the quality of Ophioordyceps sinensis strain.A set of rapid,simple and economical polysaccharide extraction process was established for Ophioordyceps sinensis in Qinghai Province,and the quality of Ophioordyceps sinensis in different areas was preliminarily evaluated from the perspective of polysaccharide based on the polysaccharide yield rate,so as to provide reference for the development and utilization of Ophioordyceps sinensis in Qinghai Province in the future.This test is to adopted 11 regional Ophioordyceps sinensis strains from Zadou County(HD-1),Yushu County(HS),Qumalai County(HM),Chengduo County(HD-2),Maqin1County(LM-1),Maqin2 County(LM-2),Dari County(LD),Xinghai County(NH),Guide County(ND),Henan County(GN),Hualong County(DL),Qinghai Province respectively as the research object,to fermenting and determining the best culturing time.Taking the yield rate of HS mycelium polysaccharide as the index,five extraction processes were screened and significant influencing factors were obtained on the basis of single factor test in combination with Plackett-Burman(PB)test.Then,the extraction process parameters of polysaccharide were optimized by response surface method(BBD).Under the optimal technology,the polysaccharide of Ophioordyceps sinensis was extracted and compared in different regions of Qinghai province,and the yield of polysaccharide was used as an index to evaluate the quality of Ophioordyceps sinensis dominant fungi in different regions.The results showed that there was no significant difference in mycelium biomass of Ophioordyceps sinensis in 11 regions of Qinghai Province under the same culturing conditions(P >0.05),and the optimal culturing time was 24 d.Under the five processing conditions,the yield of polysaccharide obtained by microwave extraction was the highest,andthe optimal parameters were as follows: 80 mesh,the ratio of material-liquid was1:26g· ml-1,3 times of extraction,microwave power was 330 W,and microwave time was 4 min.Under these conditions,the yield of polysaccharide was up to 9.06%.The yield of polysaccharide from Ophioordyceps sinensis in Qinghai Province varied from 6.84% to 9.12%,and the maximum yield of HD-1 polysaccharide was 9.12%.Based on the yield of polysaccharide,HD-1 was the best quality.
