FSH and LH synthesized and secreted by the pituitary gland can both induce and maintain normal follicles,and play a key role in animal growth and development,metabolism,reproduction,lactation and lactation,and stress responses.Occurrence and sperm production regulate the reproductive process,and FSH can directly regulate bone mass,prevent bone loss,and also reduce the accumulation of feces in the body and maintain energy replacement.Therefore,further isolate the molecular regulation mechanism of FSH synthesis and secretion.RNA(circRNA)is a special new type of endogenous non-coding RNA recently discovered.More and more studies have shown that circRNA plays a key role in various physiological activities and cancer.However,does circRNA participate in FSH breakdown.In order to explore the effect of circRNA on the secretion of adenohypophysis FSH,in the early stage,through the differential expression profile analysis of rat adenohypophysis circRNA in different developmental stages,we selected a circRNA with rich expression and differential expression,named circAkap17 b according to its source for further research.In this study,the dual luciferase reporter system and transfection experiments were used to analyze the effect of the miR-7 family on post-transcriptional recombination of FSHβ;and the interaction between the differentially expressed circAkap17 b and the miR-7 family was analyzed using bioinformatics;Expression of circAkap17 b plasmid and design of siRNA to analyze the potential effect of circAkap17 b on adenohypophysis FSH secretion;dual luciferase reporter system and RIP experiments to verify the interaction between circAkap17 b and miR-7 family.The result shows:1.The miR-7 family can reduce FSHβ and inhibit the secretion of FSH in adenohypophysis of rats.Compared with other tissue indexes of rats,miR-7 has a high expression level in the pituitary of the rat,and there is a significant difference;meanwhile,miR-7 is lowly expressed in the pituitary of the mature rat.The dual luciferase reporter system and the miR-7 transfection experiment proved that miR-7 inhibited the secretion of FSH in adenohypophysis of rats.2.CircAkap17 b can be used as miR-7 molecular sponge to secrete FSH in rat adenohypophysis.Genomic comparisons showed that the antisense strand of Akap17 b gene on chromosome X of circAkap17 b mice was identified by using fluorescence in situ crossover technique(FISH),electrophoresis,and RT-qPCR.It proves that circAkap17 b and miR-7 have an overlapping relationship,and rescue experiments also show that circAkap17 b can replace the secretion of rat adenohypophysis FSH through miR-7.In summary,this study analyzed the differential expression of circAkap17 b through the circAkap17b-miR-7-FSH signaling pathway and implanted the replication of the rat adenohypophysis FSH,and analyzed the way in which the circRNA in the rat pituitary participated in replication and replication.To provide theoretical support to analyze the preliminary biological mechanism of pituitary hormone secretion.
