Estrogen receptor(ER)have two classical subtypes which is ERαand ERβ,and distributed in various tissues and organs in the body, playing a regulatory role by binding to the estrogen of target organ. Pituitary is the most important endocrine organ of the body's, secreting various of hormones, such as PRL, FSH,LH and so on, as well as one of the target tissue of the ER.The relationship between ER and the adenohypophysis endocrine has always been one of the focus of research. In order to explore the distribution of ERαand ERβin the adenohypophysis and the relationship with hormone secretion in different physiological period. In this experiment by using different methods of single-marker immunohistochemical SP method and MaxVision double labeling to locate ERα,ERβ, PRL, FSH,LH and coexist of any two in the adenohypophysis of goat which during the estrous cycle, pregnancy and lactation. Access to the following major findings:1. The distribution of ERα,ERβ,PRL,FSH and LH in the goat adenohypophysis during estrous cycle were mainly in the chromophil cells of pars distalis in the pituitary, PRL located in the acidophilic cell,FSH and LH located in the hasophilic cell cells,ERαand ERβare distributed in the two. The expression of ERαconsistent with PRL and LH, and higher than ERβ(P<0.05), and FSH gradually increased(P<0.05).2. The distribution of ERα,ERβ,PRL,FSH and LH in the goat adenohypophysis during pregnant.Consistent with the estrous cycle. The expression of ERαconsistent with PRL, and ERβin contrary; ERβhigher than ERα(P<0.01) in the early and meddle pregnancy, ERαand ERβwas no significant difference in the late(p> 0.05).3. ERαand ERβwas coexistence respectively with PRL, FSH and LH in the goat adenohypophysis during estrous cycle. The total number of co-existence cells of ERαwith PRL, FSH and LH is greater than ERβ. ERαco-exist with PRL was highest in the proestrus, and with LH and FSH was highest in the metestrous.4. ERαand ERβwas co-existence respectively with PRL,FSH and LH in the goat adenohypophysis during pregnant and compare to lactation. The coexistence cells of ERαwith the PRL and LH was gradually increased, ERαand ERβwith FSH is opposite. the coexistence of ERαwith PRL, FSH and LH higher than ERβin peak period of lactation.Conclusions: (1) PRL located in the acidophilic cell, FSH and LH located in the hasophilic cell cells ,ERαand ERβare distributed in the two. (2) During estrous cycle The expression of ERαconsistent with PRL. Suggest the feedback function of estrogen on adenohypophysis reproductive related hormones secretion is mainly mediated by ERα. (3) The relative expression of ERβhigher than ERα.Suggest estrogen modulation of adenohypophysis hormone secretion mainly through ERβin the early and meddle pregnancy and collaborative with ERαin the late pregnancy. (4) ERαand ERβcan participate the immune response of E2 in the cells of PRL, FSH and LH in the adenohypophysis of goat in various physiological periods, and the relationship between ERαand reproductive hormones more closer.
