| Selenium(Se),as an essential trace element of the human body,has multiple biological effects.In order to solve the problem of selenium deficiency worldwide,selenium supplementation is necessary.This experiment takes jujube as the research object,cultivates selenium-rich jujube by injecting selenium-rich nutrient solution,and studies the characteristics and morphology of selenium-rich jujube by adopting selenium-rich technology.Concluded as follow:1.This experiment used the HG-AFS method to determine the total selenium content of selenium-enriched jujube.The results showed that the total selenium content of jujube leaves was always higher than that of jujube,and the total selenium content of jujube leaves reached 0.8914 mg/kg.The dry-fresh ratio of selenium-rich jujube was 45.73%,and the selenium content of jujube powder was always higher than that of fresh jujube.The highest total selenium content in jujube powder and fresh jujube was 0.7029 mg/kg and 0.6211 mg/kg,respectively,which were80.83% and 79.28% higher than that in the blank group.Therefore,selenium-enriched jujube cultivated by injecting an appropriate amount of selenium nutrient solution can be used as an effective selenium supplement product.2.The experiment selected ultra-pure water,HCl,NaOH,and Tris-HCl as extractants to study the contents of selenium in different dissolved states in Se-rich jujube.The results showed that there were four kinds of dissolved selenium distribution rules: alkali-soluble state> water-soluble state> Tris-HCl dissolved state>acid-soluble state.This shows that the selenium-enriched jujube has the strongest selenium migration and transformation ability under the alkaline condition.3.The optimum extraction conditions for soluble protein and soluble polysaccharide in selenium-enriched jujube were obtained by single factor experiments.Under the optimal extraction conditions,the maximum contents of soluble proteins and polysaccharides in selenium-rich jujube were 6.157 g/kg and71.58 g/kg,respectively,and the maximum contents of protein selenium and polysaccharides were 0.0357 mg/kg and 0.1622 mg/kg,respectively.The results showed that the ability of polysaccharide to bind selenium was stronger than that of protein.4.The biological activity of selenium-rich jujube was measured by UV method.The results showed the maximum reducing power of selenium polysaccharide and selenoprotein was 88.13% and 95.62% respectively,and the highest DPPH radical scavenging rate was 89.25% and 98.75% respectively.The results showed that the biological activity of polysaccharide selenium was higher than that of protein selenium.5.Quantitative analysis of various trace elements in selenium-enriched jujube by ICP-MS was carried out to study the biological stress of each element.Studies have shown that,with the gradual increase in the amount of selenium,selenium has a significant synergistic effect on the content of Zn,Mn,Cu,Ca,Fe,V,Mo,and Co;and selenium has significant effects on heavy metal elements such as As,Cd,Hg,and Pb.Antagonism.6.Qualitative and quantitative analysis of organic selenium and inorganic selenium in selenium-rich jujube by HPLC-ESI-MS.The results showed that Se-Met was mainly present in selenium-enriched jujube with the highest content of 16.91 μg /kg,and inorganic selenium was not present.It shows that jujube can transform inorganic selenium into organic selenium for human body absorption and utilization. |
