| Chicken gout disease is a kind of nutritional metabolic disorder disease,which is often caused by excessive uric acid production or excretion disorder in chicken body,resulting in excessive accumulation of uric acid in the blood,resulting in a large amount of uric acid deposition in the body.Clinically,it is characterized by emaciation,weakness,joint enlargement,dyskinesia and stone excretion.At present,the primary diagnosis of chicken gout is mainly through clinical manifestations and pathological changes.Therefore,in this study,diclofenac sodium was used to establish the gout pathological model of adult laying hens to determine the critical value of uric acid when chicken gout occurs.With the help of the rapid diagnosis principle of human gout,and then through the uric acid dry chemistry test,the rapid and accurate diagnosis of chicken gout was developed uric acid dry chemical test strip,and clinical application effect evaluation.The main results are as follows:1.Establishment of pathological model of gout in laying hensIn order to establish the pathological model of gout in laying hens,the chickens of group 1,2 and 3 were fed the basal diet with diclofenac sodium,and the chickens of group 4 was the control group.According to the changes of serum uric acid and pathological changes,the critical value of uric acid in gout was determined.The results showed that the mental,appetite,feces,egg production and other clinical manifestations were normal,and no pathological changes were found in all organs in group 4.The serum uric acid content was 127.83~183.06μmol/L.When the uric acid content of groups 1,2 and 3 was more than 211μmol/L on the 1th day of the experiment,the mild gout symptoms were showed,with the increase of drug dosage and the prolongation of medication time,water or stone gray thin feces chickens wer excreted,weight and egg production declined,even stopped production and appeard death.On autopsy,uric acid deposits were found in the kidney,liver,heart,pericardium,mesenteric and abdominal fat,and obvious visceral gout lesions were found,among which the 3 group was the most serious.Therefore,the critical value of uric acid was set as 200μmol/L.2.Development of uric acid dry chemical test stripAccording to the reaction principle of uric acid dry chemical test strip,the dipping solution of uric acid dry chemical test strip was prepared.The suitable ratio of reagent was 1:1.It could be stored for 3 months at 4~8℃.It had good detection linearity and good repeatability between 50~5600μmol/L.The structure of uric acid dry chemical test strip was determined.It was divided into two layers:one was support layer(plastic hard base plate:5.8×0.5cm),the other was reagent reaction layer(treated and fixed all reagents needed for reaction with sample:1.8×0.5cm).When the uric acid dry chemical test strip was used to detect the sample,the sample volume was 15μL,and the reading was the most accurate within 10~20 mins.the stability test showed that the sample can be stored for 33 days at 4~8℃with good specificity,repeatability and sensitivity.According to the detection results of uric acid in the pathological model of gout in laying hens and uric acid standard stock solution with concentration of 50~5600μmol/L,the standard colorimetric card of uric acid was determined,and the value of uric acid was set to be 0,200,500,1000,2000umol/L.3.Evaluation of the detection effect of uric acid dry chemical test stripIn order to evaluate the efficacy of the uric acid dry chemistry test paper of the clinical testing,The uric acid content of 60 serum samples collected by gout pathological model and 38 serum samples of dead chickens with kidney swelling were detected by enzyme-linked immunosorbent assay and uric acid dry chemical test striprespectively.The results showed that the square of correlation coefficient was r~2=0.9998 and r~2=0.9830,respectively,two kinds of detection method had good correlation,it proved that uric acid dry chemistry test paper had good clinical application effect of article,and could be further promotion and application in clinic. |
