As a wild medicinal fungus,Phellinus igniarius is rich in polysaccharides,flavonoids,triterpenoids,sterols,cellulase and other active substances,and operate the function of regulating immunity.At present,the research on P.igniarius mainly focused on the artificial cultivation and fermentation of mycelium,and pharmacological effect.While the research on fruiting body mainly focused on the analysis of polysaccharide and proteoglycan.Separation and screening and the optimization of solid cultivation conditions of wild strains of P.igniarius in Changbai Mountain Area were carried on in this research.Optimization of the liquid fermentation culture conditions conducive to the accumulation of polyphenols in the mycelia of P.igniarius and the anti-inflammatory effect of the active components of P.igniarius based on the rat inflammatory model was also analyzed.Firstly,the solid culture conditions of P.igniariuswere determined by using tissue separation method.The culture medium for the separation and purification of P.igniarius was PDA,the growth time of 15 days,the culture temperature of 26-30?and the water content of 60-65%.The medium of the second-class strain was wheat granule medium.The optimum temperature for the growth of fruiting body was not less than 15?at night,not more than 26?during the day,and the best culture section was wood block of oak.Secondly,in order to solve the shortcomings of harsh conditions,low biological efficiency and difficulty in achieving large-scale production in the cultivation of mulberry segment,the deep fermentation technology of fungi was explored.The medium was optimized using the L9(3~4)orthogonal experiment method.The liquid fermentation culture conditions of P.igniariuswas investigated through the single-factor experiment,and the liquid fermentation process that was conducive to the accumulation of polyphenols in P.igniariusmycelium was determined.The results indicated that the culture medium was consisted of 2.5%corn flour+20 g glucose,2%peptone,0.10%potassium dihydrogen phosphate and 0.2%magnesium sulfate.Under the initial condition on PH 5.0 of culture medium,30?of culture temperature,200 r/min of the shaker speed,the inoculation volume of 20%,100 mL of the sample loading volume and 72 h of the culture period,the highest polyphenol content was obtained from the mycelium P.igniariusand its yield was 1.58%.Finally,the anti-inflammatory experiments using polysaccharide active substance extracted from P.igniarius were carried out in the inflammatory rats.The mechanism of anti-inflammatory pharmacological action of P.igniarius was also discussed.The experimental results demonstrated that trehalose dihydrate played a part in anti-inflammatory effect,but the effect was dose-dependent.The thermal and mechanical stimulation of the left paw of the inflammatory rats infused with 10 mg/mL trehalose solution increased significantly compared with the normal saline.Compared with normal saline,the inflammatory rats that were perfused with 100 mg/mL trehalose solution showed different changes to different stimuli on the right paw.were Both left and right paw mechanical stimulation of the inflammatory rats perfused with 50 mg/mL trehalose solution were significantly increased and the anti-inflammatory activity was the best comparing with the rats with the normal saline..
