Deoxynivalenol is one of the most serious mycotoxin contamination in crops and animal feeds.When livestock and poultry consume feed contaminated with DON,it will not only cause intestinal damage but also cause toxicity of liver,kidney and other immune organs.In serious cases,it will cause vomiting,decrease food intake and decrease production performance of livestock and poultry.At the same time,it can cause serious losses to livestock and poultry breeding industry.Liver is an important detoxification organ of broilers and the most important target organ of the vomiting toxin in the body of livestock and poultry.Because of that,there have been many studies on the liver damage or liver toxicity caused by DON.Lactobacillus is the most widely used green additive,which can not only regulate the intestinal health,but also has the antioxidant effect.Our lab early experiment has proved that lactobacillus JM113 with high activity.therefore,we choose lactobacilli JM113 so as to affect the liver cells through anti-oxidation,autophagy and apoptosis gene expression.We expect to use lactobacillus efficiently to alleviate the liver toxicity of broiler chickens caused by dietary intake of DON,so as to achieve more efficient utilization of feed.Exp.1 Effects of lactobacillus on hepatotoxicity in broilers induced by food intake contaminated with DONThe experiment was carried out in a single factor design.120 AA broiler chickens were randomly divided into 4 treatments with 6 replicates for each treatment and 5 chickens for each treatment.The four treatment groups were:1)control group(CON)fed basal diet;2)the vomiting toxin group(DON)added 5 mg/kg of vomiting toxin to the basal diet;3)lactobacillus group(JM113)added 1×10~9 CFU/Kg JM113 to the basal diet.4)the mixed group(J+D)added5 mg/kg of vomiting toxin and 1×10~9 CFU/kg JM113 to the basal diet.Fasting blood collection and slaughter sampling were conducted at 21 days of age to determine relevant indicators.The test results showed that 1)the addition of vomiting toxin and lactobacillus had no effect on liver weight and liver index(P>0.05).2)compared with the other three groups,the vomiting toxin group had significant tumor-like changes in the liver tissue.3)Alkaline phosphatase(ALP)decreased in the DON group,while ALP increased in the J+D group compared with that in the DON group(P<0.05).4)The expression of the inflammatory factor IL-1 gene in the DON group was higher than that in the other three groups(P<0.05).The results of this experiment showed that the vomiting toxin could cause some damage to the liver of broilers at the age of 21 days,and lactobacillus JM113 added into the feed could relieve the liver damage caused by the vomiting toxin in some way.Exp.2 Mechanisms of lactobacilli on hepatotoxicity in broilers induced by dietary ingestion of DONThe experiment collect the broilers’liver at the age of 21 and detect the expression of oxidative stress related genes(Nrf2,HO-1),cell apoptosis related gene(Caspase3,Caspase9,Bcl2 and Bax)and autophagy related gene(LC3,ATG5,Beclin1,Dynein,m TOR)of the livers by RT-q PCR technology and determine the enzymes and materials related to oxidative stress by ultraviolet colorimetry.The experiment results showed that:1)Nrf2 in the DON group was significantly up-regulated,but the mixed group with lactobacilli was decreased compared with the DON group(P<0.05).2)the MDA level of the DON group was significantly lower than that of the other three groups(P<0.05).3)the expression of ATG5 in the DON group was significantly higher than that in the control group,at the same time,the MDA in the J+D group was lower than that in the DON group,but higher than that in the control group(P<0.05).4)the expression level of Caspase9 in the DON group was significantly increased(P<0.05).Meanwhile,it was decreased when the two groups were added together,but not significantly(P>0.05).The results of this experiment indicated that lactobacillus had certain effects on anti-oxidation,autophagy and apoptosis in alleviating the effects of vomiting toxin on broiler liver toxicity.Conclusion:lactobacillus can alleviate liver injury and liver toxicity caused by vomiting toxin in broilers to a certain extent,and it also has certain manifestations in antioxidant,apoptosis and autophagy.
