Synthetic wheat is created by artificial hybridization of Aegilops(Aegilops tauschii)and tetraploid wheat.As the intermediate of cultivated and wild species,bread wheat can directly hybridize with it,so as to obtain beneficial wild gene resources and achieve the purpose of new variety cultivation and variety improvement.QTL analysis on flag leaf,plant height and spike of hybrid progenies of cultivated wheat and synthetic wheat is helpful to carry out molecular marker assisted breeding with wild excellent gene resources.In this study,two F7 RIL populations were constructed by crossing bread wheat Xinong389,synthetic wheat KU39 and KU98,named S and J contain 136 lines and 152 lines respectively.The F7 families were used as mapping populations and 55 K wheat SNP array as typing method.Two high-density genetic maps are constructed to analyze the segregation distortion region.Through two years of phenotypic investigation and statistics,QTL analysis was carried out on the characters of flag leaf length,width,flag leaf area,plant height and spike length ect.The main results are as follows:1.Two high-density genetic maps covering 21 chromosomes were constructed.The genetic map of S population consisted of 3820 SNP markers,including 34 linkage groups,with a genome-wide length of 9785.08 c M and an average marker density of 2.56 c M.The number of genomic markers A,B and D were 1278,1321 and 1221.The genetic map of J population consists of 32 linkage groups containing 4711 SNP markers,with a total length of 10408.43 c M and an average marker density of 2.21 c M.The number of genomic markers in A,B and D were 1468,1463 and 1780.2.The genetic map of S population has 1011 segregation distortion markers,accounting for 28.82% of the total markers.Among them,686 markers were distortion towards the female parent and 415 markers were distortion towards the male parent.There were 55 segregation distortion regions with a total length of 1050.90 c M,which was 10.83% of the total length of the genetic map and distributed on 17 chromosomes except 1B,2A,5B and 6B.The genetic map of the J population has 1399 segregation distortion markers,accounting for 29.70% of the total markers,of which 374 and 1025 were distortion towards the paternal and maternal parents.There were 60 segregating regions with a total length of 1168.024 c M,which was 11.48% of the total length of the genetic map and distributed on 17 chromosomes except 1B,5B,7A and 7B.3.A total of 178 QTLs were detected on 21 chromosomes in the association population over two years,of which 70 were detected in the S population and 108 in the J population.There were 18,21,12 and 17 QTLs related to flag leaf length,width,area and leaf sheath length,16,15 and 19 QTLs related to plant height,top first internode length and spike length of neck,and 15,21 and 24 QTLs related to panicle length,spike width and chaff hardness.4.Among the populations,15 QTLs were detectable in the two years,they were q FLW-J-5A.1,q PH-S-6A.1,q TFIL-S-2D.1,q TFIL-J-1D.1,q NLS-J-2D.1,q NLS-J-7A.1,q SL-J-2D.1,q SL-J-5A.1,q SW-S-2D.1,q SW-S-6B.1,q SW-J-5A.1,q CH-S-2D.1,q CH-S-5A.1,q CH-J-2A.1,q CH-J-5A.1.5.In S and J populations,q FLW-S-6D.1 and q FLW-J-6D.1 related to flag leaf width contained the common marker AX-109158832,q SW-S-5A.1 and q SW-J-5A.1 related to spike width contained the common marker AX-108792246;q CH-S-5A.1 and q CH-J-5A.1 related to chaff hardness contained the common marker AX-108792246.
