There is a large and complex microbial population in the intestines of rodents.After a long period of adaptive evolution,a complex and dynamically balanced system has been formed in the intestine of the host.With the in-depth study of intestinal microbes,people have discovered that many factors can have an impact on intestinal microbes.And the host diet will generate strong selection pressure in the intestine,and its impact on the intestinal microbial community structure is currently a hot research topic.Gansu zokor(Eospalax cansus)belongs to the subfamily Myospalacinae of Spalacinae,and it is a typical rodent living underground.Previous studies on the intestinal tract of Gansu zokor mainly focused on exploring the adaptability of the digestive tract to the environment from the perspective of morphophysiology,while ignoring the role of “the second set of genomes” intestinal microbes of animal in the adaptation process of Gansu zokor.The study aims to reveal the characteristics of intestinal flora and metabolites of Gansu zokor during food changes,as well as the shared core flora in the intestines of females and offspring of Gansu zokor through 16 S r RNA sequencing and metabolomics technology.To provide a basis for the study of intestinal microbes in the ecological adaptability of underground rodents.The main results of this study are as follows:1.Cecal bacterial diversity of Gansu zokor under wild and artificial feeding conditionsUnder wild and artificial feeding conditions,the dominant bacteria were both Firmicutes and Bacteroidetes,but there were significant differences in abundance.Under artificial feeding conditions,the cecal bacterial diversity and structure of Mongolian pine,Chinese pine,and spruce were similar.They all had significant differences with alfalfa and wild groups.2.Cecal bacterial diversity of parental and filial generation of Gansu zokorThe dominant bacteria in cecum between the parental and filial generation of Gansu zokor were Firmicutes,Bacteroidetes,and Proteobacteria.But the abundance of Firmicutes and Proteobacteria was significantly different between the two groups.Alpha diversity analysis showed that the cecal bacterial diversity of Gansu zokor parental generation was significantly higher than that of filial generation.Beta diversity analysis showed that there were significant differences in cecal bacterial composition and structure between parental and filial generation.3.Metabolomics of the cecum contents of Gansu zokor based on food source changesUsing LC-MS technology to carry out a preliminary study on metabolites of the cecum contents of Gansu zokor under the change of food source,we found that the metabolic spectrum of the cecum contents of Gansu zokor changed significantly when the food source changed.The study found that Biocytin,D-biotin,Tryptophanol,Homovanillin,4-Hydroxyphenylpyruvic acid,Dehydroepiandrosterone,androsterone,Cortisol,and Cortolone were higher in Mongolian pine group,Gentisic acid and Tyrosol were higher in alfalfa group.Spearman correlation analysis of these metabolites with the cecal bacteria on the family level of Gansu zokor showed that the changes of metabolites were related to the anundance of intestinal microbes.
