In this study,Ipomoea aquatica Forsk.was used as the material,and hydropnic experiments conducted by six different monochromatic with red and blue compound LED light source.Adding the different proportions of green light in the early screening the best red and blue compound light ratio.Finally,in the use of different light quality of water spinach on the impact of light on the resistance of white rust of water spinach,when under the different light quality.seeking to achieve high yield and good quality of water spinach growth and postharvest freshkeeping storage period of the best mass ratio of light,provides the theory basis for production of quality control.The main research results are as follows:1.The effects of different LED light quality on growth,quality and nitrate ofwater spinach.Studies have shown that the monochromatic light compared with the CK(white light),red is the most beneficial to swamp cabbage plant height,leaf soluble protein of the stems of Vc,improve,reduce the leaf nitrate,and blue B the most favorable enlargement of stem,leaf and root of the accumulation of soluble sugar,green G good swamp cabbage plant height,to a certain extent,contributed to the swamp cabbage root soluble protein increased,stem Vc content.Compared with CK(white light),the water spinach with red and blue composite light 80R/20B had the highest height and fresh weight,which was beneficial to the accumulation of soluble sugar,soluble protein and Vc in leaf,stem and root.It is most beneficial to decrease nitrate content in stem and leaf of spinach and increase nitrate reductase activity.2.The effect of different green light ratio on growth,quality and nitrate content of water spinach.With red and blue composite light 80R/20B as the background light,The results showed that the plant height,stem diameter and fresh weight of water spinach were higher than CK(white light).The content of soluble sugar,soluble protein and Vc in leaves was higher than that in CK(white light),among which the red,blue and green mixed light 72R/18B/10G was the highest,while CK(white light)was the lowest.During the harvest period,the nitrate content in stem and leaf treated with different light quality was significantly lower than CK(white light),among which 72R/18B/10G treatment had the best reduction effect.Plant height,stem diameter,root volume,root surface area and average root diameter were the largest under 72R/18B/10G treatment.It was concluded that adding 10%green light to red and blue composite light was the most beneficial to the growth and quality improvement of spinach,and also the most beneficial to the decrease of nitrate content in stem and leaves.Therefore,the ratio of 72R/18B/10G water spinach could be used as the best allocation of water spinach,and reveal the potential value of green light and its role in water spinach production.3.The effects of different LED light quality on the incidence of water spinach white rust and its quality and related enzyme activity.The results showed that the incidence of white rust in spinach was lower than CK,and blue light treatment was the best.Blue light treatment of swamp cabbage leaf phenylalanine ammonia enzyme solution(phenylalanine ammonia lyase,PAL)activity is significantly higher than other processing;The activity of Superoxide dismutase(SOD)in different parts of stem was significantly higher than that in other treatments.The PAL activity of the upper stem and root of water spinach was significantly higher than that of other treatments.SOD activity of leaves and roots was also significantly higher than that of other treatments.The correlation analysis showed that PAL activity was positively correlated with the morbidity of water spinach leaves and negatively correlated with PAL activity in middle stem.4.The effect of different LED quality on the change of NRA,homologous acquisition and PCR amplification of NIA1 gene.The results of physiological experiments were verified from the molecular point of view.The study showed that compared with CK(white light),the relative expression of NIA1 gene was increased by R treatment compared with red light,and the relative expression of NIA1 gene by green light G and blue light B was lower than CK.The red and blue composite light treatments all increased the activity of nitrate reductase and the expression of NIA1 gene.Under the red,blue and green composite light treatment,72R/18B/10G treateient significantly increase.d the relative expression of NIA1 gene,while 64R/16B/20G was lower than CK.Reverse transcription PCR wa;s used to sequence the homologous PCR products of spinach leave's with arabidopsis,Japanese petunia and sweet potato.Qrtpcr analysis showed that the relative expression of NIA1 gene was the.largest under the treatament of red,blue and green composite light 72R/18B/10G,which indicated that the expression of NIA1 gene could be promoted under this light quality ratio and the accumulation of nitrate could be reduced.The relationship between the variation of NRA caused by light quality and the expression of NIA1 gene during nitrate assimilation was revealed,and the conclusion of physiological experiment was verified.
