Lipid metabolism plays important role in animal growth,development and reproduction.Previous studies reported that cidec promotes unilocular development of lipid droplet and stimulates lipid storage in multiple types of cells,indicating cidec is a crucial regulator in lipid metabolism.Recent work showed cidec has two isoforms,cidec-l and cidec-s.cidec-l have 10 more animo acids in N-ternimal compared to cidec-s.However,the functions of two iosoforms are largely unknown.In this study,the expression profile of two isoforms were detected in Bama pig.The data showed that cidec-l is main isoform in liver and small intestine,but cidec-s is dominant isoform in muscle and adipose tissue.By fasting or high-fat diet treatment,we found that the expression changes of two isoforms are closely related to blood and muscle triglyceride(TG)alteration.Through comparing the gene expressions and TG levels of DLY and Bama pig,it suggests that cidec-l tends to accelerate lipid secretion,but cidec-s tends to promote lipid storage,implying two isoforms have different function.Study in vitro proved this hypothesis.cidec-s stimulated lipid storage in C2C12 muscle cells,but cidec-l promoted lipid secretion in Hep G2 liver cells.Altogether,our work revealed that cidec plays a bi-function in lipid storage and secretion by tissue-specific two isoforms.It provides a new target to regulate lipid metabolism of blood and tissues.
