Pork quality is an economically important trait and the key index for pig breeding at present.With the improvement of people’s living standards, pork quality traits from the beginning of the blind pursuit of lean meat to now pay more attention to quality. Intramuscular fat content is an extremely important indicator of meat quality, and ability of the liver metabolism of fat deposition levels.Cell death-inducing dFF45-like effector c (CIDEC),also known as Fat-specific protein of 27 kD (Fsp27) in mus, is related to fat deposition and palys an important role in the process of fat metabolism.Since CIDEC is a high degree of homology (82.8%) in sus and mice. In the firststage, mice were choosen as the research object. Methods used RT-PCR, Co-IP,Mass Spectrometry and so on.screening at proteins interacting with CIDEC in liver and adipose at fasting,high fat diet or normal diet.The results are as follow.1. In the opposite process of the fat deposition(high fat diet) and fat mobilization(fast process), the expression of CIDEC and the content of triglyceride (TG) in liver was significantly increased.It suggests that CIDEC might play an important factor role in fat deposition and fat mobilization.2. In our research,we used Co-IP and mass spectrometry technology to capture and screen proteins interacting with CIDEC in liver and adipose, then we analyse proteins interacting with CIDEC in aspects of protein function, subcellular localization, GO, KEGG functions and enrichment functions by DAVID website. We found in different nutrient and different organizationsconditions,the function of CIDECwas change.The second stage will use Bama miniature pigs as experimental subject, we attempts confirm whether proteins interacting with CIDEC can be established in pigs. According to the results of mass spectrometry, we selected eight significant different of proteins interacting with CIDEC andconstructed expression vectors can prepare to the next yeast two-hybrid method.
