Molecular Cytogenetic Study On The Germplasm Line Of Cold-hardy Perennial Tritelytrigia

Abstract/Summary:
In this study,the perennial cold-hardy tritelytrigia(perennial tritelytrigia hereafter),the hybrid offspring of obtained from the cross between the octoploid tritelytrigia"Wheatgrass 8"(2n=8X=56,AABBDDEE)and Thinopyrum intermedium,was used as the experimental lines.The morphological investigation,molecular cytogenetic detection,and chromosome configuration analysis were used to determine the morphological characteristics and genomic composition of the perennial cold-hardy wheatgrass germplasm.The objective was to provide the foundation for cold hardiness and perennial characteristics research on perennial cold-hardy.The results of morphological investigation showed that 21 perennial tritelytrigia were resistant to cold and regenerated after harvest.They could return to green in April at the following year,and their morphological characteristics tended to be stable.The plant height of these materials was more than 1m,and the tiller number was 11-102,which was significantly more than common wheat.19-7111 and 19-414156 had the highest seed setting rate,exceeding 80%.The cytological analysis showed that the chromosome number of 21 materials tended to 42,except for 19-42529356 that was between 43-45 chromosomes.Meiosis observation of pollen mother cells found that19-42529356 had the most unvalents.19-41213355,19-414153173,19-412133204,19-7111,19-425293125,19-425299128,and 19-425299129 had the most ring bivalents.19-425299121,19-425299124,and 19-425299126 had most rod bivalents.The results of molecular markers and GISH showed that the chromosome number of ABD group in perennial tritelytrigia was between 12 and 21.19-414156 and 19-7111that had the better seed setting carried 16 and 12 ABD chromosomes,respectively.The alien chromosome of wheat in perennial tritelytrigia was mainly 3-10 St chromosomes,3-20 J chromosomes,and J~s group was less.In common tritelytrigia of SX184,SX186and SX188,the chromosomes number of ABD groups was 39,42,and 33,respectively.The alien chromosome number was 4-6 St chromosomes,0-5 J chromosomes,2-6 Js chromosomes and the translocations were occurred between ABD chromosome and alien chromosome.The St group specific primers found that St chromosomes in perennial tritelytrigia were mainly 4St and 5St.St and J chromosome count of common tritelytrigia(the control)was significantly less than perennial tritelytrigia.Therefore,it revealed that cold resistance and perennial traits were related to the chromosome group of St and J.
