Platycladus orientalis is a local product of China,which is distributed in most parts of the country.Cuttage is the key technology to realize clone breeding and improved seed breeding of trees,but the induction technology system of Platycladus orientalis cuttings rooting is not perfect,and the changes of nutrients such as soluble sugar,enzyme activities such as POD,PPO,IAAO,IAA,ABA,GA3,ZR and other endogenous hormones changes in cuttings rooting are not known.The lack of understanding of differential proteomics in the process of adventitious root development of Platycladus orientalis has become a bottleneck restricting clone breeding and fast propagation of fine varieties of Platycladus orientalis.This study by Beijing Academy of Forestry and Pomology Sciences using superior resources Platycladus orientalis 'Liye',2 years of cutting seedlings,2 years of grafting cuttings as object,The changes of soluble sugar,soluble protein,POD,PPO,IAAO and other enzymes,as well as the contents of IAA,ABA,GA3,ZR and other endogenous hormones in the process of advadvent root generation were studied;iTRAQ and LC-MS were used to study the differential proteomics in the rooting process of cypress lateralis 1.The main results are as follows:1.The content of soluble sugar,soluble protein,POD,PPO and IAAO activities were related to the occurrence of root.Before cutting(0d),soluble sugar content,IAAO and POD activity had the greatest influence on callus formation(swelling or adventitious root formation),followed by soluble protein and least influence on PPO activity.During the induction period of root primordium(40d),soluble sugar content and IAAO activity had the greatest influence on rooting rate,followed by soluble protein and POD,while PPO activity had the least influence.Therefore,during the rooting process,the contents of soluble sugar,soluble protein,IAAO and POD activities in cuttings were closely related to rooting,while the activities of PPO were not closely related to rooting.2.In the process of adventitious root development,IAA and ABA are the key factors affecting adventitious root development(or callus induction).During the callus formation period,the contents of GA3,IAA and ZR in cuttings and grafted cuttings increased to varying degrees,while the ABA content decreased.During the induction period,the ZR and ABA contents of the three kinds of cuttings decreased,while the contents of IAA and GA3 increased,and the cuttings and grafted cuttings decreased.The content of IAA,ZR and ABA of the three types of cuttlet decreased in the whole from the time of adv.root extension.Therefore,it can be concluded that endogenous IAA significantly promoted the rooting of cuttles,with emphasis on promoting callus formation or adventitious roots.ABA content is not the main reason to inhibit rooting,but to assist in rooting.3.The different proteome of basal cortex of cuttings in different stages of pruning was analyzed by the combination of iTRAQ and LC-MS.During the four stages of cutting,callus formation,callus swelling and adventitious root formation,4946 different proteins were identified from the cortex of cuttings,among which there were 1,863 up-regulated proteins and 3083 down-regulated proteins.The proteins closely related to rooting were selected from the obtained differential proteins:auxin IAA hydrolase,ferritin NADP+oxidoreductase,pseudoglutamate deoxidase,cytochrome b6 and photosystems CP43.Through the comparative analysis of GO functional annotation and KEGG database,the main functions of these differential proteins in the process of advanced-root generation include cellular processes,metabolic processes,biological regulation,stimulus reactions,catalytic activities and protein binding.The metabolic pathways involved are mainly the processing of genetic information,energy,carbohydrate,amino acid metabolism,lipid metabolism and biosynthesis of other secondary metabolites.
