Differenced Study Of Berry Anthocyanin Accumulation Between Vitis Vinifera And Hybrid Of Vitis Vinifera × Vitis Amurensis In Red Wine Grapes

In this study,16 Vitis Vinifera and hybrid of Vitis Vinifera × Vitis Amurensis red wine grape varieties were selected as materials.The content of 5 monomer anthocyanins in grape fruits were detected by HPLC.The expression levels of anthocyanin synthesis related structural genes and regulatory genes were detected by quantitative real-time PCR(qRT-PCR).To explore the differences in fruit quality and anthocyanin accumulation of different wine grape varieties,which providing a theoretical basis for the introduction or breeding of red grape varieties in the Eastern of Helan Mountain.The results showed as followed:1.The results of the study show that there are significant differences in the fruit quality of different wine grape varieties.At the ripening stage,the 100-grain weight of Vitis Vinifera ’Cabernet Franc 623’ and ’Cabernet Sauvignon 170’ was smaller,the total soluble sugar content of the fruit of’Marselan’ was higher,’Pinot Noir’,’Cabernet Sauvignon 169’ and ’Marselan’ accumulated more titrable acid and tannin,total phenol and anthocyanin content respectively.Compared with Vitis Vinifera grapes,Vitis Vinifera×Vitis Amurensis grapes show high sugar and high anthocyanin properties,it can be found that:the 100-grain weight of Vitis Vinifera × Vitis Amurensis ’Beixi’ was smaller,the total soluble sugar content of ’Beixin’ was significantly higher than other varieties,’Beihong’ fruit has the highest titratable acid and phenolic content.2.The results of significant differences were found among different wine grape varieties,regarding the content of 5 monomeric anthocyanins.During the harvest period,the contents of Cy-3-O-G,Pt-3-O-G and Dp-3-O-G were the highest in Vitis Vinifera ’Sangiovese’,the Pn-3-O-G content of ’Marselan’ fruit was higher,and the content of Mv-3-O-G was higher in ’Syrah’ fruit.The contents of Pt-3-O-G fruit of ’Beihong’ was higher,and the content of Mv-3-O-G was higher in’Beichun’ grape.It was showed that the content of monomer anthocyanins in Vitis Vinifera grapes was higher than that in Vitis Vinifera × Vitis Amurensis grapes.3.In terms of enzyme activity related to anthocyanin synthesis,compared with Vitis Vinifera grapes,the PAL,C4H and 4CL enzyme activities of Vitis Vinifera×Vitis Amurensis grapes were higher during harvest,and the enzyme activities of ’Beihong’ grape fruits were the highest.4.Regarding gene expression related to anthocyanin synthesis,there are significant differences among different wine grape varieties.At harvest,the relative expression level of the fruit of Vitis Vinifera grape ’Marselan’ was significantly higher than that of other varieties;the relative expression of VvC4H was the highest in grape ’Sangiovese’.The relative expression level of VvF3 ’H in grapes of Vitis Vinifera×Vitis Amurensis grapes was significantly lower than that of Vitis Vinifera grapes,and the relative expression of Vitis Vinifera grape ’Syrah’ VvF3’H was the highest.The relative expression level of VvF3 ’5’H was lower in Vitis Vinifera and Vitis Vinifera × Vitis Amurensis grapes,and compared with the control,the grape varieties of Vitis Vinifera×Vitis Amurensis grapes except ’Beihong’ were down-regulated.The relative expression level of VvUFGT was highest in the ’Beixi’ and ’Cabernet Franc 623’.Compared with Vitis Vinifera grapes,the relative expression of VvMybAl was higher in the grapes of Vitis Vinifera×Vitis Amurensis grapes,and the gene expressions of ’Xinbeichun’ and’Beixi ’were the highest.Therefore,through the gene expression of VvPAL,VvF3’H,VvF3’5’H and VvUFGT,different species of grapes regulate the related enzymes in the synthesis of PAL,C4H and 4CL,and affect the accumulation of 5 monomeric anthocyanins in fruits.The accumulation of anthocyanins in the fruits of Vitis Vinifera grape ’Marselan’ and Vitis Vinifera×Vitis Amurensis grape ’Beihong’ was significantly higher than that of other experimental varieties,under the conditions of this experiment in the Eastern of Helan Mountain in Ningxia.
