Artemisia anomala,commonly known as Liuyueshuang in China,is a perennial herbaceous plant of Artemisia species belonging to Compositae family in china,and widely used as traditional medical ingredients for hundreds of years.Many studies showed that A.anomala possess bioactive effects including clearing heat,relieving summer heat and breaking blood circulation.However,the tea made from the A.anomala harvested had a poor taste and the tea soup was cloudy and easily discolored and deteriorated.Therefore,people usually store the A.anomala for three years after harvesting.After being stored for three years,the color of the tea soup is orange and clear,and not easy to change color and overnight metamorphism.In order to scientifically clarify the reason why A.anomala needs to be stored for three years,this topic uses the composition and distribution of chemical components in A.anomala as an entry point to clarify the changes in ingredients during storage.Therefore,in this study,A.anomala in different storage years was used as the research material to analyze the composition and content of protein,amino acids,total flavonoids,total phenols,and volatile components,and to compare the differences in composition and content.1.The protein content of A.anomala was determined by spectrophotometry,and the composition and content of amino acids of flowers were evaluated by automatic amino acid analyzer.The results showed that A.anomala contained high protein content,and there were differences among different parts and storage years.The content of protein in flower(192.19 mg·g-1)was significantly higher than that in the peduncle(62.70 mg·g-1)and stem(33.11 mg·g-1).However,with the increase of storage years,protein content decreased significantly in the third year.There are 16 kinds of amino acids in the flowers of different storage years,the total amino acid content of flowers stored for three years was highest(91.12 mg·g-1).In addition,the content of flavor amino acids increased year by year,indicating that the storage time is conducive to the improvement of the taste.2.The contents of total flavonoids and phenols of A.anomala were detected,the ability of scavenging DPPH· and ABTS+ radicals and the total reducing power were determined.The results showed that A.anomala contained high total flavonoids and phenols content,and had good antioxidant capacity.The content of total flavonoids and phenols in flower were significantly higher than that in the peduncle and stem,while the content of n-butanol extract of flower was the highest.With the increase of storage time,the content of total flavonoids and total phenols and antioxidant capacities of A.anomala decreased.Moreover,there was significant positive correlation between the antioxidant capacities and the contents of total flavonoids and total phenols,which indicated that the total flavonoids and total phenols in A.anomala had better antioxidant activities,and the main active part was n-butanol extract.3.The volatile compositions of A.anomala were determined by HS-SPME-GC-MS.The results indicated that the best extraction conditions of SPME were 1.2 g of sample weight,70 of ?heating temperature and 60 min of adsorption time.According to GC-MS analysis,45 kinds of volatile compounds were identified from A.anomala,including 45 kinds of flowers,42 kinds of pedicels,40 kinds of stems,and there were 40 common components among them.Among them,(-)-caryophyllene oxide,cineole and(+)-borneol acetate are the main components in flowers and pedicels,while ?-farnesene,humulene epoxide ii and(-)-caryophyllene oxide are the main components in stems with low content.With the increase of storage years,the content of most terpenoids decreased,while the contents of(+)-borneol acetate and?(-)-?-thujone?(+)-?-thujone and ?-cyperone with special flavor increased year by year.The results of principal component analysis showed that the volatile components of pedicel and stem were similar and not easily affected by the storage time,but the components of flower were slightly different from the former two,and the volatile components were significantly different between different storage years.
