According to the survey,in many places of Beijing,jujube trees and lily plants appeared mosaic,leaf-rolling,yellowing,the typical symptoms of viral diseases,affecting the economic and ornamental values.In this study,deep sequencing technology was conducted to identify the virus(es)in jujube trees and lily plants,which supplied a foundation for the prevention and treatment of viral disease in jujube and lily.The main experimental results were as follows:1.Four unreported viruses and a new isolate were found in jujube:(1)Jujube yellow mottle-associated virus(JYMaV),in the genus Emaravirus of the family Fimoviridae,had six RNA strands with each strand encoding the RdRp,glycoprotein precursor,nucleocapsid protein,movement ptotein,P5 protein,P6 protein respectively.(2)Jujube mosaic-associated badnavirus(JaBV),in the genus Badnavirus of the family Caulimoviridae.The circular DNA genome of the virus had four open reading frames,with each encoding the unknown protein,virion-associated protein,polymerase,unknown ptotein respectively.(3)Chinese jujube leafroll-associated virus(CJLRaV),in the genus Ampelovirus of Closteroviridae.The virus is a single-stranded RNA virus,and it has 7 open reading frames.(4)Apple stem grooving virus(ASGV),belonging to Betaflexiviridae family and Capillovirus genus.This RNA viruse has two open reading frames encoding the polymerase and movement ptotein.(5)Chinese jujube virus F(CJVF),belonging to the Fabavirus genus in the family Secoviridae.The virus is a bipartite virus;RNA1 has an open reading frame,encoding the RNA helicase and reverse transcriptase;RNA2 has an open reading frame that encodes the movement,large coat protein and small coat protein.2.Four unreported viruses were found associated with the lily plants:(1)lychnis mottle virus(LycMoV),belonging to the Secoviridae family(unassigned genus).The virus has two RNA strands;RNA1 has an open reading frame,encoding the protease cofactor,RNA helicase,genome-linked viral protein,protease and polymerase;RNA2 has an open reading fram that encoded the movement ptotein,large coat protein and small coat protein.(2)Lily yellow vein virus(LYVV),belonging to Caulimoviridae family and Rosadnavirus genus.The virus has a circular DNA genome and part of the sequence was obtained including the movement ptotein,capsid protein and putative protein.(3)Lily western yellows virus(LPV),belonging to Luteoviridae family and Polerovirus genus.The virus has a single-strand RNA,and four open reading frames were obtained:ORF0 encoded the P0 protein,ORF1 encoded the RdRp P1 protein,and ORF2 encoded the RdRp p1-p2 fusion protein.The other parts had not been completed.(4)Lily endosomal virus(LEV),belonging to Endornaviridae family and Endornavirus genus.The virus had a single-stranded RNA,and part of the genome sequence were obtained:the unknown region,the glycosyltransferase,the Hel and RdRp,other parts hadnot been completed.
