Longiflorum lily is a perennial bulbous flower of lily family.It is one kind of important ornamental lilies and an important parent for intersection cultivars.However,due to the serious virus infection caused by long-term asexual reproduction,the quality of bulbs was greatly reduced.Therefore,its virus detection and detoxification have certain practical significance on the production of high-quality lily bulbs.In this study,Longiflorum lily ’ white fox ’ was used as experimental material,and its scales were treated with water at different temperatures and sterilized according to normal in vitro culture.Then the bulbs and scales produced by in vitro culture were subcultured with different sucrose concentrations.The viruses carried on the obtained plantlets by the in vitro culture were detected by RT-PCR technology,so as to analyze the effect of different water temperature treatments on the sterilization and detoxification of lily scales.The main results are as follows :(1)The effects of different water temperatures on the sterilization and survival of lily scales were significantly different : the polluted rates of 35℃,45℃,55℃ and 65℃ for 15 min were 39%,20%,6% and 0%,respectively,and the survival rates were 35.5%,70.5%,11.5 % and 0 %,respectively;The contaminated rates of 35℃,45℃,55℃ and 65℃ for30 min were 32.5 %,14.5%,0% and 1% respectively,and the survival rates were 43.5%,75.5%,0% and 0% respectively.According to the results,the treatment of 45℃for 30 min for lily scale sterilization and scale survival effect is ideal.(2)Different sucrose concentrations had different effects on bulb enlargement and differentiation in subculture.Sucrose concentration of 0 g/L,30 g/L,50 g/L,70 g/L for bulb expansion coefficient were 1.36,2.01,2.1,2.69,the differentiation rate were 28.67%,92.67%,78%,24.67%.Combined with the results of bulb expansion coefficient,differentiation rate and dry matter content,it was preliminarily shown that the sucrose concentration was 50 g/L,it was the most suitable for the growth and development of bulblets.(3)The detoxification effects of different water temperatures on Lily symptomless virus(LSV),Lily mottle virus(LMoV)and Cucumber mosaic virus(CMV)were significantly different.The detoxification rates at 35℃,45℃ and 55℃ for 15 min were 0%,46.67% and 100%,respectively;The detoxification rates at 35℃ and 45℃ for 30 min were 3.33% and 73.33%,respectively.According to the results,the treatment at 55℃ for15 min had the best detoxification effect,but the death rate of scales was above 85%.Therefore,the detoxification effect of LSV,LMoV and CMV at 45℃ for 30 min was ideal.In summary,the treatment of Longiflorum lily ‘ white fox ’ scales at 45℃ for 30 min was ideal for sterilization of lily scales,survival rate of scales and detoxification of three main viruses.Sucrose concentration of 50 g/L was most suitable for small bulb growth.
