By analyzing the transcriptome data of full flowering stage of soybean cytoplasmic male sterile line and its homotypic maintainer line,the mechanism of soybean cytoplasmic male sterility was clarified,and the theoretical basis for the utilization of soybean cytoplasmic male sterility in sterile seed production was provided.Using cytoplasmic male sterile line jlcms116a and its homotypic maintainer line jlcms116b as materials,carbohydrate,soluble protein,endogenous hormones and transcriptome sequencing of the two materials at full flowering stage were analyzed.The transcriptome data of all materials produced 40.53gb of sequence data,a total of 1866 genes were obtained,and 229 differential genes were screened out between male sterile line and maintainer line.1.GO annotation analysis showed that 157(68.6%)of 229 differential genes were annotated into 42 functional categories,including 19 biological classifications(BP),12 cellular component classifications(CC)and 11 molecular functional classifications(MF).2.KEGG enrichment showed that the major pathways of DEGs enrichment between CMS and maintainer lines were.pentose and glucuronic acid mutual transformation pathway(ko00040),starch and sucrose metabolism(ko00500),plant pathogen interaction(ko04626),sphingolipid metabolism(ko00600)and ether lipid metabolism(ko00565)Xie(ko00500),sphingolipid metabolism(ko00600)and ether lipid metabolism(ko00565)were related to fertility.3.Eleven genes closely related to fertility were screened by KEGG enrichment analysis.These genes mainly encode pectin lyase,polygalacturonase,pectinase,pectinase inhibitor,pectinesterase and endoglucanase.4.The analysis of five hormones in the flower bud of male sterile line and maintainer line showed that the auxin and cytokinin in of male sterile line were lower than that of its homotype maintainer line,and ethylene content was higher than that of its homotype maintainer line.5.The analysis of soluble sugar,soluble protein and starch content in flower bud of male sterile line and maintainer line showed that the content of soluble sugar,starch and soluble protein were lower than that of maintainer line.
