Whether as a model animal for scientific research or as a protein meat to meet People's daily demand,chicken genetics has a long history and good prospects for contributing new knowledge and global food.In order to adapt to modern commercial production,how to efficiently obtain the male germ cells of the cock with good quality has become one of the key technologies.Embryonic stem cells(ESCs)has the potential of differentiation into all kinds of cells,including the male germ cell.The differentiation of ESCs into male germ cells is coordinated and regulated by many factors.Therefore,only by clarifying the regulatory mechanism of these factors and establishing an effective induced differentiation system in vitro,can the generation efficiency of male germ cells be fundamentally improved.MicroRNAs(miRNAs),as a gene regulatory factor,is involved in numerous fields.With the increasing discovery of miRNAs,many of them have been identified as important regulators in poultry reproduction.We have analyzed the RNA-seq data and screened out miR-302d which is differentiated and negative during the differentiation of ESCs(Embryonic stem cells)into SSCs(spermatogonia stem cells)induced by RA in vitro.LncRNA-341 acting as a ceRNAs that can competitively bind to miR-302d to decrease the targeted binding of miR-302d and Tle4.This study contributes to the gap of miRNAs in regulating the differentiation of avian male germ cells,and provides the theoretical support to establish efficient culture system of male germ cells differentiation and improve production efficiency of male germ cells.
