Objective: In order to investigate the express characteristics of stem cell in testis issues, the histologic characteristics of testis issues and distribution regularity of fos-related antigen Oct4,Nanog,C-kit andβ1-integrin of Spermatogonial Stem Cells (SSCs)in various livestock animals at different ages were studied.Methods: The present study was undertaken to investigate the tissue structure of holstein cows fetus at 2.5, 4 months and 1, 2 years old after birth; White Goat at 45, 75 days and 4 months; fetal hybrid piglet from Duroc,Landrace and Large White at 60 days and half a year after birth by HE staining and immunohistochemistry.Results: In the development progress from sexual differentiation to birth investigated by histology observation, the Primordial germ cells (PGCs)at early foetus stage assembled very close to be obvious group shape cell colony which was separated by Sertoli cell. With increase of the age,the spindle-shaped myoid cell was distributed outside of seminiferous tubules,and spaced the cell groups,formed seminiferous tubules,and the atret-lumens swelling gradually, and finally formed the lacune. With the swell of lumens, part of spermospore close to seminiferous epitheliubaselemman gradually formed sperm.Immunohistochemistry to make clear that Oct4,Nanog,C-kit andβ1-integrin were in different testis tissue. SSCs had more content in testicular issue of smaller fetal aged livestock, and the number of masculine cell was bigger. With growth of fetal age, the number of positive cell relatively decreased. But after the birth of livestock , the SSCs in testicular tissue still exists, but relatively less content.Among the total, (1) the expression of Oct4 in various livestock testicular tissue was the most widespred, and Nanog expression trend as the same as Oct4, but the relative quantity decreases. Among them, in 2.5, 4 months cattle foetus, 45 day-old sheep foetus'testicular tissue, positive cells expressed in seminiferous tubule and interstitial cell; 75 day-old sheep foetus and 60 day-old pigs foetus, expressed in a circular distribution around seminiferous tubule, and relatively less content in interstitial tissue cell; 1 year post partum cattle and 4 months post partum sheep, positive cells were distributed in testicular issue Basement member of seminiferous tubule, and less or none in interstitial cell; 2 years after birth cattle and half a year after birth pig, positively expression were a little and none in distributed in testicular issue Basement member of seminiferous tubule.(2)C - kit andβ1 - integrin was in membrane expression. Among them, in 45 day-old sheep foetus testicular tissue, positive cells was distributed island-shaped in most cell membrane of both seminiferous tuble cells group and interstitial cells.; 75 day-old sheep foetus positive cells in testicular issue were relatively less, and membrane expressed in substance part of convoluted tubles; in 60 day-old pig foetus testicular tissue, positive expressed in seminiferous tubule basilemma cells, but not in interstitial cells around tubule; in 2.5, 4 month-old cattle foetus testicular tissue, musculine cells were obvious clumps linked together; in 4 months post partum sheep, 1 year post partum cattle testicular tissue, positive cells were relatively less, generally membrane expressed in basilemma of seminiferous tubule basilemma cells, accidentally seen in interstitial cells around tubule. In half a year birth pig's testicular tissue, C - kit positive cells decrease relatively, and mostly found in cell membrane of interstitial cells around tubule, andβ1 - integrin were expressed in both seminiferous tubule basilemma and interstitial cells around tubule. In two years post partum cattle's testicular tissue, positive expression was less found, occasional sporadic membrane expression in seminiferous tubule, and a small quantity expression.in interstitial cells around tubule。Conclusions: SSCs had more content in testis issue of smaller aged livestock, and the number of masculine cell was bigger. With fetal age growth, the number of masculine cell decreased. But after the birth of livestock , the SSCs in testicular tissue still exists, but content relatively decreased obviously.
