In order to determine the protective effect of schisandrin on animal myocardial hypertrophy injury,this paper carried out artificial model tests on myocardial hypertrophy injury in vivo and in vitro after pretreatment of rat schisandrin with rat cardiomyocytes,intragastric test animals and target animals:Schisandrin protects against norepinephrine-induced cardiomyocyte hypertrophy injury.Rat H9C2cardiomyocytes were cultured in vitro,5?g/mL captopril positive control group,5?g/m L,10?g/mL,20?g/mL low,middle and high schisandrin group,intervention for 48 h,adding 10~-66 mol/L norepinephrine successfully induced myocardial hypertrophy in 48 hours.Observe cell morphology,detect myocardial cell surface area and protein/DNA ratio,determine changes in BNP,IL-6,TNF-?,IL-10,etc.;The test results show that the schisandrin medium-dose group can significantly reduce mast cell surface area,protein/DNA ratio and BNP,IL-6,TNF-?content(P<0.05),and increase IL-10content(P<0.05),Improve the survival rate of myocardial cells(P<0.05),reduce the apoptosis rate(P<0.05),restore the mitochondrial membrane potential,down-regulate the expression of BAX,up-regulate the expression of Bcl-2 protein,and effectively relieve myocardial hypertrophy.Experimental study on the protective effect of Schisandrin on adriamycin hydrochloride-induced myocardial hypertrophy.Pre-gavage SD male rats,10 mg/kg/d captopril positive control group,9 mg/kg/d,18 mg/kg/d,36 mg/kg/d low,middle and high schisandrin drug group,The blank control group was gavaged with equal volume of normal saline for a total of 21 days,intraperitoneal injection of 2 mL/kg doxorubicin hydrochloride according to the body weight of rats for 6 weeks can successfully induce myocardial hypertrophy.Observe the behavioral changes of rats,detect changes in heart weight index,left ventricular wall thickness,and determine the content of BNP and inflammatory factors;The test results show:Schisandrin medium dose can significantly reduce heart weight index,left ventricular wall thickness and BNP,IL-6,TNF-?content(P<0.05),reduce the concentration of RBC,HGB,RDW,MVP,pathological observation of myocardial fiber hypertrophy and other lesions are reduced,myocardial apoptosis rate is reduced,BAX expression is down-regulated,Bcl-2 expression is up-regulated,relieve myocardial hypertrophy.Experimental study on the protective effect of schisandrin on myocardial hypertrophy of target animals caused by abdominal aortic constriction.Pre-gavage male dogs,3 mg/kg/d captopril positive control,6 mg/kg/d schisandrin group,a total of 14 days later,Abdominal aortic constriction surgery caused canine myocardial hypertrophy in 4 weeks,and the control group was given an equal volume of normal saline.Observe changes in dog behavior,detect changes in heart rate HR,systolic blood pressure SBP,heart weight index,left ventricular wall thickness,determine BNP and inflammatory factors.The dog test results showed that Schisandrin can reduce the white blood cell concentration and significantly reduce the content of IL-6(P<0.05).Conclusion,schisandrin has a certain protective effect on myocardial cell hypertrophy in vivo and in vitro,Mainly by reducing the content of BNP,IL-6,TNF-?,down-regulating BAX protein,up-regulating Bcl-2 protein,protect cardiomyocytes;at the same time,it reduces the content of IL-6,the target animal of myocardial hypertrophy injury,and reduces the inflammatory injury.The test results provide a reference for further exploring the protective mechanism of schisandrin on myocardial hypertrophy,it provides a certain experimental basis for the research and development of schisandrin for the prevention and treatment of canine myocardial hypertrophy.
