The mechanism of onion poisoning in puppies is that the sulfur-containing compounds in Onions are hydrolyzed to thiosulfonate after mastication,and then the thiosulfonate is decomposed into many disulfide compounds,among which the n-propyl disulfide will reduce the activity of glucose-6-phosphate dehydrogenase(G6PD)in red blood cells.Glucose-6-phosphate dehydrogenase protects hemoglobin from oxidative degeneration.If the activity of glucose-6-phosphate dehydrogenase is reduced,the hemoglobin will be oxidized and coagulated,which leads to the rupture of red blood cells to form the Heinz body,which leads to anemia in puppies.In this paper,the effects of physiological and biochemical indexes on onion poisoning in puppies and the differences between the physiological and biochemical indexes of Chinese garden puppies and golden retriever puppies were discussed,which provided a reliable theoretical basis for the diagnosis and treatment of onion poisoning in puppies.In this study,six Chinese garden puppies and six golden retrievers were selected and divided into group A and group B.The self-control before and after feeding Onions and the inter-group control of group A and group B were adopted.The puppies were fed Onions30g/kg body weight,for 3 consecutive days.In the week before feeding Onions and 1 d,3 d,6 d,9 d,16 d,18 d and 24 d after feeding Onions,the puppy's blood was collected for blood routine and biochemical examination to observe the changes of physiological and biochemical indicators after onion poisoning.The test results are as follows:1.Changes of physiological indexes: On the first day after the onion was fed,Heinz body appeared,and the surface of red blood cells was protuberant;On the 6th day after the onion was fed,the cell membrane of red blood cell ruptured most seriously,and the phenomenon of dissolution appeared;On the 9th day after the onion was fed,the breakdown of red blood cells was less than that on the 6th day;On the 18 th day after the onion was fed,the membrane surface of a few red cells appeared protuberance,and no dissolution of the membrane of redcells was found;On the 24 th day of onion feeding,a few red blood cells were deformed,and there was no protuberance or dissolution of the membrane of red blood cells.RBC,HCT and HGB were all decreased after the onion was fed,and decreased to the lowest value on the 6th day after the onion was fed.The difference was significant(P< 0.01);The mean corpuscular volume(MCV),leukocyte(WBC)and mean hemoglobin content(MCH)were all increased after onion was fed,and increased to the highest value on the 6th day after onion was fed,which was significantly different from that before onion(P< 0.01);Platelets(PLT)gradually increased after onion feeding,and reached the highest value on the 3rd day after onion feeding.Compared with that before onion feeding,the difference was very significant(P<0.01).PLT always fluctuated within the normal range during the whole experiment;Average red blood cell hemoglobin concentration(MCHC)values gradually rising slowly after feeding the onion,3 days after feeding onion peak,compared with feeding before the onion,there is no difference(P> 0.05),then gradually reduced,13 days after feeding the onion,dropped to the lowest,compared with feeding before the onion,difference was significant(P< 0.01),and then gradually increase to feeding value before the Onions.2.Changes in biochemical indicators:Total protein(ALB)decreased gradually after onion feeding and fluctuated in the normal range throughout the experiment;The total protein(TP)value gradually increased after onion feeding,and reached the highest value on the third day after onion feeding.Compared with that before onion feeding,the difference was not significant(P> 0.05),and then gradually decreased.During the whole experiment,the total protein value fluctuated within the normal range;The value of urea nitrogen(BUN)dropped to the lowest value on the 1st day after feeding onion,which was significantly different from that before feeding onion(P< 0.01),The difference was significant(P< 0.05),and then decreased,It began to rise on the 13 th day after the onion was fed,and the value of urea nitrogen fluctuated within the normal range throughout the test;Alanine aminotransferase(ALT)value gradually increased after onion feeding,and reached the highest value on the 3rd day after onion feeding.Compared with that before onion feeding,the difference was extremely significant(P< 0.01),then it gradually decreased,and began to rise on the 13 th day after onion feeding.During the whole experiment,alanine aminotransferase alwaysfluctuated within the normal range;The total bilirubin(TBIL)value increased gradually after onion feeding,and reached the highest value on the 3rd day after onion feeding.Compared with that before onion feeding,the difference was extremely significant(P< 0.01),and then it gradually decreased,and fell to the normal range on the 6th day after onion feeding;Creatinine(CRE)value gradually decreased after onion feeding,and began to rise on the 6th day after onion feeding,and then gradually decreased again.During the whole experiment,the value of creatinine fluctuated within the normal range,and the difference was not significant compared with that before onion feeding(P> 0.05).3.Comparison of physiological and biochemical indexes of Chinese garden puppies and Golden retriever puppies: The changes of physiological and biochemical indexes were not related to puppy breed after onion poisoning.
