Probiotics,a beneficial active microorganism,are widely used in the fields of nutrition,health care and medicine,with the characteristics by potent biological function to protect animal organism.Currently,existing studies have reported that the biological function of inactivated probiotics is similar to viable bacteria in many aspects,moreover,inactivated probiotics is superior in the safety,stability and resistance than viable bacteria.As a major energy supply,Polysaccharide is an ideal medicated premix due to litter side effects for the body and it can prevent pathogenic microorganisms from invading the body,resist the proliferation of tumor cells and regulate the body's immunity.However,the research on immune additives is most limited to one or one kind and the combined effects of polysaccharides and probiotics are rarely reported.In this study,Hyline Chickens were used for conducting following experiment.The chickens were randomly divided into 6 groups,blank control group?BC?,cyclophosphamide model group?CY??80mg/kg.BW?,Enteromorpha Polysaccharide group?CY+EP??100mg/kg.BW?,low?1x108CFU/mL?,medium?1x109CFU/mL?,high(1x1010CFU/mL)dose of inactivated Bacillus Subtilis and Enteromorpha Polysaccharide combination group?CY+IVC+EP?.First,immunosuppression model was established.The control group is injected with saline and others 5 groups are treated with the same amount of cyclophosphamide by intramuscular injection.Second,each group randomly selected from the same number of chicken after irrigating for 7d,14d,21d,collecting heart blood to measure blood physiological parameters,IgM,IgG,IL-2,IFN-?and Newcastle disease?ND?antibody titers of chickens.Then,The thymus,spleen and bursa of fabricius were collected to detect the immune organ index,the content of SIgA in duodenum lotions,AvBD9 and AvBD13 mRNA expression in the organization.The main test results are as follows:Effect of Enteromorpha Polysaccharide combined with inactivated Bacillus Subtilis on immune organ index of immunosuppressed chicken.compared with the blank control group,the indexes of immune organs in the cyclophosphamide group were all reduced,Moreover,during the test period,thymus index and bursa index were significantly different from the blank control group?P<0.01?.Compared with the cyclophosphamide group,the immune organ index increased to different degrees after administration of Enteromorpha Polysaccharides and inactivation of Bacillus Subtilis in immunosuppressive chicken,and the combination of Enteromorpha Polysaccharides and inactivation of Bacillus Subtilis had a more significant effect on immune organ index relief than single administration of Enteromorpha Polysaccharides.The results showed that cyclophosphamide could cause the decrease of immune organ index in chicken,while Enteromorpha Polysaccharide and inactivated Bacillus Subtilis could alleviate the decrease of immune organ index,and the combined effect of the two was better.Effect of Enteromorpha Polysaccharide combined with inactivated Bacillus Subtilis on serum antibody titerof immunosuppressed chicken.compared with the blank control group,the serum antibody titer of the cyclophosphamide group was reduced,the lavage after 7d,14d,and the blank control group significant difference?P<0.05?,gastric 21d,and the blank control group difference was significant?P<0.01?.Compared with the cyclophosphamide group,when Enteromorpha Polysaccharide and inactivation of Bacillus Subtilis were administered to immunosuppressive chicken,the titer of antibody against NDV was significantly increased?P<0.01?,and the effect of Enteromorpha Polysaccharide and inactivation of Bacillus Subtilis combined group on the reduction of antibody titer was better than that of Enteromorpha Polysaccharide alone.The results showed that cyclophosphamide could reduce the titer of serum antibody in chicks,while Enteromorpha Polysaccharide and inactivated Bacillus Subtilis could alleviate the titer decrease,and the combined effect of the two was stronger.Effects of Enteromorpha Polysaccharide combined with inactivated Bacillus Subtilis on serum immunoglobulin and cytokines in immunosuppressive chicken.compared with the blank control group,serum immunoglobulin?IgG and IgM?and cytokines?IL-2 and IFN-??were decreased in the cyclophosphamide group.Compared with cyclophosphamide group,immunosuppressive chicken to infuse suit Enteromorpha Polysaccharides and inactivate Bacillus Subtilis,ease in serum immunoglobulin?IgG and IgM?and cytokines?IL-2 and IFN-??content decreased,and in addition to the IFN-?,Enteromorpha Polysaccharides and inactivate Bacillus Subtilis joint effect of IgG,IgM and relief IL-2 levels lower than single irrigation Enteromorpha Polysaccharides is better.The results showed that cyclophosphamide could reduce the contents of immunoglobulin and cytokines in the serum of chicken,while Enteromorpha Polysaccharides and inactivated Bacillus Subtilis could alleviate the decrease,and the combined effect of IgG,IgM and IL-2 was stronger.Effects of Enteromorpha Polysaccharide combined with inactivated Bacillus Subtilis on SIgA content in duodenal lotion of immunosuppressive chicken.compared with the blank control group,SIgA content in duodenal lotion of cyclophosphamide group was significantly reduced?P<0.05?.Compared with the cyclophosphamide group,administration of Enteromorpha Polysaccharide and inactivation of Bacillus Subtilis to immunosuppressive chicken could alleviate the reduction of SIgA content in the duodenal lotion of immunosuppressive chicken,and the effect of Enteromorpha Polysaccharide and inactivation of Bacillus Subtilis combined group on the reduction of SIgA content was more significant than that of Enteromorpha Polysaccharide alone.The results showed that cyclophosphamide could reduce the content of SIgA in duodenal lotion of chicken,while Enteromorpha Polysaccharide and inactivated Bacillus Subtilis could alleviate the decrease of the content,and the combined effect of the two was stronger.Effects of Enteromorpha Polysaccharide combined with inactivated Bacillus Subtilis on the expression of AvBD9 and AvBD13 mRNA in immunosuppressive chick tissues.compared with the blank control group,the expressions of AvBD9 and AvBD13 mRNA in the chick tissues of the cyclophosphamide group were significantly decreased?P<0.01?.Compared with the cyclophosphamide group,the expressions of AvBD9 and AvBD13 mRNA in the immunosuppressive chicken increased to different degrees after administration of Enteromorpha Polysaccharide and inactivation of Bacillus Subtilis,and the expression of AvBD13 mRNA increased with the increase of inactivated Bacillus Subtilis concentration in the combined group.The results showed that cyclophosphamide could reduce the expression of AvBD9 and AvBD13 mRNA in chick tissues,while Enteromorpha Polysaccharide and inactivated Bacillus Subtilis could alleviate the decreased expression,and the combined effect of the two was stronger.In summary,Enteromorpha Polysaccharide combined with inactivated Bacillus Subtilis can by improving immunosuppression chicken immune organ index,serum antibody titer,immunoglobulin and cytokine levels in serum,duodenum lotions SIgA,AvBD9 and AvBD13mRNA relative expression in the content and organization,such as ways to give play to the role of immune regulation.
