Transposons are sequences that can be moved on chromosomal DNA and are widely found in organisms.The Phyllostachys edulis accounted for 59% of the whole genome,and its distribution and function have great research value.The distribution of exons and introns in each transposon family in the Phyllostachys edulis genome was analyzed by the repeatmasker software.1.61% of the transposons were distributed in the gene,and the exon and intron regions accounted for 11.57% and 88.43,respectively.%.Through analysis,it was found that the length of the transposon distributed in the gene region was shorter than the length of the transposon distributed in the intergenic region,and the length of the transposon of the distributed exon was shorter than the length of the intron transposon,which may be related to The host genome is involved in the transposon inhibition effect.The DNA,RC and SINE transposons prefer to insert intron regions,while the LINE class transposon prefers to insert exons and introns,and the LTR prefers to be inserted outside the gene.Six introns containing a transposon sequence were selected to analyze the effect of the transposon in the intron on gene expression.The transposon-containing and transposon-containing intron sequences were amplified and introduced into the GUS gene.The expression of GUS gene was analyzed by the transient expression system and quantitative PCR technique,and the forward and reverse introns were found.The effect of insertion on the gene is different,and the overall expression enhances the expression of the gene.The transposon inserted into the intron region slightly inhibits the enhancement of the intron.Two exons with a transposon sequence were selected to analyze the effect of transposon on gene expression in the exon.Amplify the CDS sequence containing the transposon and the transposon,and the sequence containing only the transposon,insert the plant genetic expression vector PCM1301,transfer it to Arabidopsis,and find that the gene containing the transposon is relatively free of transposition.The gene of the sub-genome shows that the expression has more advantages.Ex0949 competes with the homologous gene and Ex4969 competes with the downstream gene.The gene deletion is lost after the transposon is removed,which is expressed as homologous gene and downstream.The expression of the gene is up-regulated,and the single transposon has no effect on the downstream gene.A transposon Ex4969,which may cause variable shear,was also found,the insertion of which is coupled with variable shear.The above experiments show that the transposon inside the gene will have an effect on the expression of the gene.These effects may be beneficial to gene expression,such as transposons in exons,which may inhibit gene expression,such as introns.Transposon in the middle,but the specific transposon function needs specific analysis.
