Hunan,as one of the cradles of peony,has been cultivated for thousands of years.In the long-term natural selection,the local peony in hunan has formed the excellent characteristics of high temperature and high humidity.However,due to the abundance of rain and pests in peony in hunan,the yield and seed yield were low,and the oil was mainly used for oil,the relative lack of ornamental resources,the development of peony in hunan is stagnant.In this experiment,18 varieties of peony in hunan and 11 morphological characters were used as experimental materials,ISSR molecular marker technique was used to study the genetic diversity and genetic relationship of 29 samples,It provides a theoretical basis for sorting out the peony germplasm resources in hunan and determined the direction for the breeding of hybrid and fine varieties.The main results are as follows:(1)Using PopGen32 software analysis:The peony PPB was 95.73%,Na was 1.9573,Ne was 1.3427,H was 0.2202 and I was 0.3546.The PPB of Hunan native peony is 81.10%.The results show that the genetic diversity of peony resources in hunan is high.(2)By analyzing the cluster diagram,it is concluded that: When the valve value is 0.74,there are five main types of peonies,There are 5 varieties in the first category,including ‘Fengdanbai’、‘Fengdanxing’、‘Fengdanfen’、‘Fengdanzi’、‘Anhuifengdan’、‘Fengdan’,its colors are all white except for ‘Fengdanfen’and ‘Fengdanzi’ which are pink and purple,The flowers are all single-flowered,among them,’Anhui Fengdan’ is clustered with Hunan’s indigenous Fengdan,indicating that the genetic relationship between them is relatively close;There are 5 varieties in the second category,including ‘Fengdanlv’、 ‘Fengdanyu’、‘Fengdanling’、‘Fengdanwu’、‘Yangshanmudan’,its colors are all white and The flowers are all single-flowered;There are 8 varieties in the third category,including ‘Ningxianghong’、‘Baoqinghong’、‘Baoqingfen’、‘Lijiaxiang’、‘Roufurong’、‘Zixiuqiu’、‘Xiangxiuqiu’、 ‘Yongshunfen’,among them,‘Roufurong’ and Hunan native peony are clustered into one category,which shows that their kinship is closer;There are 7 varieties in the fourth category,including ‘Sichuanzixiuqiu’、 ‘Sichuanzifen’、 ‘Pengzhouzi’、‘Luoyanghong’、‘Taipinghong’、‘Xiangziban’、‘Honglian’,among them,the‘Xiangziban’ and the Northwest Purple Peony’s‘Honglian’are clustered.It shows that the relationship between Hunan native purple spotted peony and Northwest purple spotted peony is relatively close;There are 3varieties in the fourth category,including‘Fenhe’、 ‘Honghaiyingbo’、‘Shushengpengyue’,The three northwestern peony and the Hunan native peony gathered farthest,indicating that their kinship is farthest.The comprehensive results showed that the genetic relationship between native peony cultivars in Hunan is closer than that of peony cultivars outside the province.The genetic differences between them are relatively large. |
