Cleistogenes songorica,a xerophytic grass,is a constructive specie and dominant specie in desert grassland.With high feeding value,cold and drought tolerance,it can be used as forage grass,eco-grass and turfgrass.Up to date,there is not yet large-scale molecular markers in C.songorica,which restrict its molecular biology and breeding progess.In this study,efficient intron length polymorphism(ILP)molecular markers were developed and testes its transferability among other species,which provided ideal molecular markers for germplasm utilization and breeding program in C.songorica.The main results were as follows:1.A total of 220,165 introns were derived from the whole genome of C.songorica.Among them,181,681 introns can be mapped on 27,314 genes,and 20 pairs of chromosomes in C.songorica.A total of 20,503 genes were annotated in GO database and 9,604 genes were annotated in KEGG database.We designed 91170 pairs of Cleistogenes songorica ILP(Cs_ILP)marker primers in this study,and 84,635 Cs_ILP markers can be mapped on chromosomes.2.Using 417 pairs of Cs_ILP markers,the genetic diversity was analyzed among 23 Cleistogenes keng populations.A total of 402 alleles were identified.The average expected heterozygosity(He)and polymorphism information content(PIC)were 0.61 and 0.56 respectively.It was found that Cs_ILP markers could distinguish different species of genus Cleistogenes and the geographical distribution of different populations.Cs_ILP markers were suitable for the phylogenicclassification and genetic diversity study of Cleistogenes keng.3.C.songorica and other 10 different species plants were used to analyze the transferability of Cs_ILP markers.It was found that the transferability of Cs_ILP markers in these species was 58.40%.The phylogenetic tree could distinguish the speciesfrom different families.It indicates that the Cs_ILP markers showed high universality in species of different families.
