Stropharia rugosoannulata Farlow is suitable for raw material cultivation.It must be covered with soil before it can produce mushrooms normally.Therefore,it is presumed that covering soil is not only to provide support for the growth of fruiting bodies of Stropharia rugosoannulata Farlow,but also because it contains microorganisms that can affect the growth of Stropharia rugosoannulata Farlow.In this paper,the microbial flora of Stropharia rugosoannulata Farlow in the substrate and soil layer were preliminarily studied by means of microbial plate separation and counting and high throughput sequencing.The cultivation of 44 hybrid strains of Stropharia rugosoannulata Farlow was studied.The main results are as follows:1.Determination by microbial plate separation count.The results showed that the total number of bacteria and actinomycetes in the substrate was in line with the exponential function,and the number of moulds was in line with the logistic growth curve.2.Research on bacterial diversity and population abundance in substrates and overburden at different times by second-generation sequencing showed that the abundance of proteobacteria,β-proteobacteria,Burkholderia,the orders of Burkholderia,the families of Burkholderia was increasing as the inoculation time was extended.Among them,the number of bacteria of the genus Burkholderia Paraburkholderia reached the highest in the mushroom stage.3.Two strains of early mushroom were selected from 44 hybridization strains.The strain has the characteristics of fast mycelial germination and early mushroom emergence.
