Bupleurum chinense DC.is the plant of the Bupleurum L.Umbelliferae,r oot as medicine in China,widely distributed in most areas of the country.At pr esent,the wild resources can not meet the demand of the market,artificial cult ivation has been carried out,but the seed of Bupleurum chinense DC.is small and less,at the same time,there are some problems,such as thick seedcoat,low germination rate,disagreement of seed germination time,irregular emergen ce and dormancy of some seeds,the development scale and productionbenefit of the artificial planting of Bupleurum chinense DC.have been seriously affect ed.This experiment is based on the seed,so as to provide some guiding signi ficance for the cultivation of Bupleurum chinense DC..In this study,the seed of Bupleurum chinense DC.was used as the materi al to determine the seed quality,cleanliness,water content and water absorptio n.Use medicament and water bath to break seed dormancy,and cultured in di fferent culture temperatures,the changes of seed germination potential,germinat ion rate,germination index,vigor index and seedling root length and seedling height were observed.On the basis of the above study,salt stress treatment of seeds and seedlin gs of Bupleurum chinense DC.,then application of ALA,the changes of seed germination potential,germination rate,germination index,vigor index and seed ling root length and seedling height,and seedling protective enzyme activity an d MDA content were observed.The effect of different explants,medium,hormone ratio,culture temperatur e and light time on callus induction of Bupleurum chinense DC.was investigat ed.The effects of different hormone ratio,sucrose concentration,light time and subculture cycle on callus growth of Bupleurum chinense DC.were investigate d.The results are as follows:1.The pure rate of seeds is 91.51%,the weight of one thousand seeds is(2.83±0.03)g and the weight after seed absorbing water is about 2.24 times more th an seed dry weight.The content of moisture of seeds is 7.3%.The pretreatme nt of Bupleurum chinense DC.seed was soaked in 0.6 mg/L 6-BA,40℃2 h,and the initial culture temperature was 15℃,after germination,it continued to grow at 20℃,which could effectively improve the germination rate and seedl ing rate of seeds.2.The appropriate concentration of ALA not only alleviated the inhibition effec t of salt solution on Seed Germination of Bupleurum chinense DC.,but also pr omoted the growth of its rhizomes.ALA had a great influence on the activitie s of protective enzymes and MDA contents in seedlings.Especially when the a ctivities of POD and MDA in salt control group and distilled water control gro up did not change significantly,ALA played a more significant role in seedlin gs.3.The callus induction rate of Bupleurum chinense DC.is the highest with the base of leaves as explants,B5+6-BA 3 mg/L+2,4-D 2.2 mg/L+KT 0.79 mg/L,25℃dark culture conditions.The callus is yellow,shining and glittering,an d the growth is good.The callus of Bupleurum chinense DC.Suitable growth B5+6 mg/L 6-BA+1 mg/L 2,4-D+1.4 mg/L KT,Sugar content 2%,25℃dark culture conditions,Subculture cycle is 28d.
