Liver injury is a pathological condition associated with a variety of liver diseases.It often exists in the occurrence of acute or chronic liver diseases and involves a wide range of diseases.Acute liver injury is a common disease in clinic.If it is not treated and treated in time,it will cause necrosis and apoptosis of liver cells in the liver tissue,which will lead to liver failure,even cirrhosis or liver cancer.Acute liver injury induced by LPS is a common experimental animal model of acute liver injury in rodents.The medicine to "Radix Bupleuri-Paeoniae Paeoniae" is a classic medicine used in the treatment of liver disease in Chinese veterinary medicine.In this study,a rat model of acute liver injury induced by LPS was established,and the effects of Bupleurum and paeony on acute liver injury were studied.1.pharmacological effects of Bupleurum and Paeonia lactiflora on LPS induced acute liver injury: a rat model of acute liver injury was established by intragastric administration of Bupleurum and Paeonia lactiflora in low,middle and high doses to rats and intraperitoneal injection of LPS.It was found that all organs index was abnormal in rats stimulated by LPS,and pathological changes were observed in liver pathology.Abnormal blood routine and hematological function were significantly increased in rats.The pretreatment of Radix Bupleuri-Radix Paeoniae Radix Paeoniae Alba was relieved of organ index,tissue lesion and hematological index.The above results show that "Radix Bupleuri-Radix Paeoniae Radix Paeoniae" can effectively alleviate the liver damage of LPS induced acute liver injury,and has certain protective effect on the liver.2.Oxidative stress and anti-inflammatory effects of drugs on the acute hepatic injury induced by LPS: Biochemical tests for oxidative stress factors and ELISA for the expression of inflammatory factors.The effect of pretreatment with "Bupleurum and Radix Paeoniae Rubra" on the upregulation of hepatic GSH-PX,SOD,and IL-10 under LPS stimulation,and downregulation of MDA,NO,iNOS,TNF-?,IL-1?,and IL-6 effect.Description: Bupleurum and white peony can effectively relieve oxidation and inflammatory stimuli.3.this study by RT-PCR,Western Blot and "Cocktail" probe drug law,from three aspects of gene expression,protein expression and enzyme activity of medicine,found that hepatic enzymes CYP1A2,CYP2C11,CYP2D6,CYP3A1,CAR and nuclear receptors PXR and AhR were inhibited by a promoting effect on liver enzyme CYP2E1 in acute liver LPS induced damage,medicine of "Chaihu Baishao" pretreatment can reduce the abnormal movements of hepatic enzymes and nuclear receptors,the medicine of Bupleurum and radix paeoniae alba on acute liver injury mechanism have a preliminary understanding.In conclusion,the drug has a good liver protective effect on acute liver injury induced by LPS,and plays a role in relieving the abnormal organ index and reducing the liver tissue lesions and maintaining the balance of blood system.It also plays the role of anti oxidative stress injury and anti-inflammatory injury.Led to the inhibition of hepatic CYP1A2,CYP2C11,CYP2D6,CYP3A1 and PXR,CAR and nuclear receptor AhR expression and activity in LPS,to promote the enzyme activity and CYP2E1 expression of hepatic drug case,the expression and activity of drug intervention of liver enzyme and nuclear receptor of "Chaihu Baishao",it plays a role in restoring the stability of liver enzyme.
