Alfalfa is an important legume forage,which has the advantages of rich in nutrition and widely ecological adaptability.It is a high-quality pasture planted in large areas in China.The contents of mineral elements Ca,Zn and Fe,flavonoids and feed quality are important indicators for evaluating the quality of alfalfa.In recent years,the research on the yield and quality of alfalfa mainly focused on the fertilization and soil conditions.There are few studies on the content of flavonoids and the feed quality in alfalfa.In this study,the flavonoids content and feed quality of233 alfalfa were determined,which provided a theoretical basis for the cultivation and utilization of alfalfa and the industrialization of grass industry.The main results are as follows:(1)In this study,we developed a high-throughput method to determine 20 kinds of flavonoids in alfalfa by ultra-high performance liquid chromatography-diode array detection-electrospray ionization-mass spectrometry(UHPLC-DAD-ESI-MS).(2)Determination of 20 flavonoids in 233 alfalfa showed that there weresignificant differences in the flavonoids of alfalfa between different materials.Based on the highest and lowest flavonoids,we screened alfalfa materials and found that the highest apigenin content was No.107 and the content was 784.19 ng/mg dry weight of the leaves and the lowest was No.1 and the content was 1.45 ng/mg dry weight of the leaves,with a difference of 540.82 times.The highest glycycoumarin content was No.31 and the content was 0.71ng/mg dry weight of the leaf and the lowest was No.1and the content was 0.15ng/mg dry weight of the leaf,with a difference of 4.73 times.The highest content of daidzin was No.1 and the content was 1.73ng/mg dry weight of the leaf and the lowest was No.19 and the content was 0.01ng/mg dry weight of leaves,with a difference of 17.30 times.The highest content of glycitin was No.133 and the content was 30.75ng/mg dry weight of leaves and the lowest was No.3 and the content was 0.1ng/mg dry weight of leaves,with a difference of 307.50 times.The highest content of Liquiritin was No.1 1.39ng/mg dry weight of leaves and the lowest was No.7 and the content was 0.03ng/mg dry weight,with a difference of59.67 times.The highest content of apigenin-7-O-glucoside was No.1 and the content was1.66ng/mg dry weight of leaves and the lowest was No.9 and the content was0.07ng/mg dry weight of leaves,with a difference of 23.71 times.The highest content of luteolin-7-O-glucoside was No.160 and the content was 7.64ng/mg dry weight of leaves and the lowest was No.13 and the content was 0.08ng/mg dry weight of leaves of,with a difference of 95.50 times.The highest content of genistin was No.117 and the content was 20.86 ng/mg dry weight of the leaves and the lowest was No.2 and the content was 0.03 ng/mg dry weight of the leaves,with a difference of695.33 times.The highest content of isoliquiritin was No.1 and the content was1.21ng/mg dry weight and the lowest was No.145 and the content was 0.01ng/mg dry weight of the leaves,with a difference of 121.00 times.The highest content of liquiritigenin was No.144 and the content was 7.67ng/mg dry weight of the leaves and the lowest is No.8 and the content was 0.07ng/mg the dry weight of leaves,with a difference of 109.57 times.The highest content of glycitein is No.14 and the content was 8.00ng/mg dry weight of leaves and the lowest is No.151 and the content was 0.05ng/mg dry weight of leaves,with a difference of 160.00 times.The highest content of naringenin was No.64 and the content was11.43ng/mg dry weight of leaves and the lowest was No.184 and the content was 0.41ng/mg dry weight of leaves,with a difference of 27.87 times.The highest content of luteolin was No.173 and the content was 40.99ng/mg dry weight of leaves and the lowest was No.148 and the content was 0.3ng/mg dry weight of leaves,with a difference of 136.63 times.The highest content of genistein was No.1 and the content was 6.07ng/mg dry weight of leaves and and the lowest was No.19 and the content was 0.04ng/mg dry weight of leaves,with a difference of 151.75 times.The highest content of isoliquiritigenin was No.144 and the content was 13.88ng/mg dry weight of leaves and the lowest is No.196 and the content was 0.06ng/mg dry weight of leaves,with a difference of231.33 times.The highest content of isorhamnetin was No.177 and the content was3.51ng/mg dry weight of leaves and the lowest was No.114 and the content was0.01ng/mg dry weight of leaves,with a difference of 351.00 times.The highest content of chrysoeriol was No.21 and the content was 67.98ng/mg dry weight of theleaves and the lowest was No.216 and the content was 1.77ng/mg dry weight of the leaves,with a difference of 38.41 times.The highest content of Medicarpin was No.1and the content was 13.57ng/mg dry weight of leaves and the lowest was No.147 and the content was of 0.2 ng/mg dry weight of the leaves,with a difference of 67.85 times.The highest content of formononetin was No.1 and the content was15.87ng/mg dry weight of leaves and the lowest was No.173 and the content was0.04ng/mg dry weight of leaves,with a difference of 396.75 times.The highest content of biochanin A was No.196 and the content was 1.76ng/mg dry weight of leaves and the lowest was No.50 and the content was 0.061ng/mg the dry weight of leaves,with a difference of 2.89 times.According to the top three levels of the compounds,39 alfalfa were screened.(3)The feed quality of 233 alfalfa were measured.The results showed that there were significant differences in alfalfa feed quality between different materials.The content of Ca was between 2.060 and 4.247%,the highest content was No.198 and the lowest No.3.The content of neutral detergent fiber was between 0.143 and19.980%,the highest content was No.150 and the lowest No.102.The content of acid detergent fiber is between 27.563 and 50.537%,the highest content was No.103 and the lowest No.80.The content of acid detergent lignin was between 0.010 and4.643%,the highest content was No.206 and the lowest No.64.The ash content of alfalfa was between 14.233 and 28.103%,the highest content was No.42 and the lowest No.3.The in vitro digestibility of dry matter was between 56.870 and87.617%,the highest content of alfalfa was No.39 and the lowest No.153.(4)Finaly,based on the content of various feed quality and flavonoids,39 alfalfa with high quality were screened from 233 alfalfa.
