Han Per sheep is a new breed of meat sheep bred by crossing Small tailed Han sheep as female parent and Dorper sheep as male parent.After breeding,the meat performance of Han Per sheep was significantly improved than that of female parent,but the reproductive and behavioral characteristics need to be measured and observed.In this paper,the reproductive data of Han Per sheep were analyzed,and the testicular tissues of Han Per sheep at different months of age were observed,and the immunohistochemistry and expression of related genes were analyzed.The daily behavior of Small-tailed Han Per sheep,Han Per sheep of different months of age and estrus Han Per sheep were analyzed,which could provide reference for the breeding and daily management of Han Per sheep.Experiment 1: The reproductive data of Han Per sheep were analyzed to provide reference for future breeding.In this experiment,the lambing number,lambing number,twin lambing rate and lambing month distribution of Han Per ewes in 1-4 self-propagating generations,different generations were compared and analyzed.The results showed that the lambing number,lambing interval and twin lamb rate of Han Per sheep were lower than those of small tail Han sheep(P < 0.05).The lambing number of Han Per sheep was proportional to the parity.The lambing concentration was from January to April,with the most lambing in March and the least in July.Experiment 2: The purpose of this experiment was to study the testicular development and the possible role of related reproductive genes in Han Per sheep,so as to provide basis for the selection of breeding rams and the determination of mating age.In this study,three Han Per sheep at birth,7 months old and 13 months old were selected and their testicular tissues were collected for HE staining,immunohistochemistry and fluorescence quantitative test.The results of HE staining showed that the testicular development of Han Per sheep accorded with the characteristics of this species.FSHR(Follicle stimulating hormone receptor),LHR(Luteinizing hormone receptor)and AR(Androgen hormone receptor)were all positive in seminiferous tubule lumen of neonatal Han Per sheep.LHR and AR werepositive in spermatogenic cells and spermatozoa of seminiferous tubule of 7 and 13 months old.FSHR,LHR and AR were expressed in testicular tissues of different months old.The expression of FSHR increased in the testicular tissues of Han Per sheep at birth and 7 months old,and decreased at 13 months old.The expression of LHR in 7-month-old and 13-month-old children was significantly higher than that in newborn children(P<0.01),and the expression of AR gene in 7-month-old children was significantly higher than that in 13-month-old children(P<0.05).The expression of LHR was basically consistent with the histochemistry results,suggesting that LHR might be related to spermatogenesis in Han Per sheep,and could be used as a candidate gene for breeding sheep.Experiment 3: This experiment analyzed the daily behavior of Han Per sheep,and provided reference for the daily management and feeding of Han Per sheep.The experimental sheep were divided into five groups: adult Small-tailed Han sheep,adult Han Per sheep,3-month-old Han Per sheep,6-month-old Han Per sheep and estrous Han Per sheep.The behavior of feeding,rumination,rest,watching and movement of experimental sheep were observed and analyzed.The results showed that the feeding performance of Han Per sheep was better than that of small tail Han sheep.The feeding frequency of adult Han Per sheep was significantly lower than that of 3-month-old and 6-month-old Han Per sheep(P<0.01),while the rumination frequency was significantly higher than that of 3-month-old and 6-month-old Han Per sheep(P<0.05).There were significant differences in rest and wait-and-see behavior between estrous Han Per sheep and non-estrous ewes.The feeding duration of 3-month-old Han Per sheep at noon was significantly higher than that of adult sheep(P<0.05).In daytime,the rest frequency of adult sheep was significantly higher than that of 3-month-old and 6-month-old sheep(P<0.05),while in night,the rumination frequency of adult sheep was significantly higher than that of3-month-old sheep(P<0.01),and the exercise frequency was significantly different(P<0.05).The observation frequencies of estrus sheep in daytime and night were higher than those of non-estrus sheep(P<0.05).The feeding of experimental sheep was concentrated in daytime,rumination and rest were concentrated in night.Conclusion: The reproductive performance of Han Per sheep is lower than that of Small-tail Han sheep,and the reproductive regularity is seasonal.The breeding performance needs to be improved.The testicular tissue development of Han Per sheep accords with the development characteristics of this breed.LHR gene mayaffect spermatogenesis and can be used as a candidate gene for breeding rams.The feeding performance of Han Per sheep was better than that of Small-tailed Han sheep,and the feeding performance of 3-month-old and 6-month-old Han Per sheep was better than that of adult sheep,but the ruminant ability was lower.Han Per sheep have obvious oestrus behavior,feeding behavior is concentrated in the daytime,rumination and rest behavior is concentrated in the night.
