The swine MHC(major histocompatibility complex)or swine leukocyte antigen(SLA)is one of the most closely-linked and the most polymorphism regions in the swine genome.And it mainly related to swine infectious diseases and vaccine-induced immune response.The SLA gene,which is one of the genetic factors that cause cell immune rejection in xenografts,mainly contains 5 highly polymorphic loci,including 3 class ? classical genes(SLA-1,SLA-2 and SLA-3)and 2 class ? functional genes(SLA-DQB1 and SLA-DRB1),In order to study the pathogenesis of various diseases to provide reliable experimental animals and clarify the relationship between SLA allele and disease,what's more,to enrich the classical experimental animals model.This study uses SPF Yorkshire and Landrace,which from Harbin veterinary research institute,CAAS and imported from Canada,as experimental object to analysis.The whole study is researched through feeding management,reproductive physiological characteristics,and the SLA gene characteristics analysis.And the main results are following:1?The suitable conditions of SPF pigs' growth and development were analyzed by monitoring the environment barrier of SPF Yorkshire and Landrace.The healthy growth of SPF pigs were ensured by different disinfection and sterilization methods.We randomly selected 5 11-month-old SPF Yorkshire,5 11-month-old SPF Landrace and their first piglets respectively to detect reproductive physiological indexes.And we selected 20 16-month-old SPF Yorkshire,24 16-month-old SPF Landrace and 20 1-month-old Yorkshire piglets and 20 1-month-old Landrace piglets to analyze the physiological and biochemical characteristics.The results showed that reproductive physiological indexes and most of physiological and biochemical indexes of blood of SPF Yorkshire and Landrace were not significant difference(P>0.05),which showed population were healthy.Individual indicators such as blood physiological indicators(GRAN,HGB,RDW,PLT,PCT)and blood biochemical indexes(ALKP,CHOL,TBIL,BUN)were significantly difference(P<0.05)among varieties,different ages and sexes,but its deviation from the normal range is small,compared with other miniature pigs,each index was no significant difference(P>0.05),and partial indicators were higher similar to people.2?SLA gene was detected by PCR-SBT and PCR-SSP,and 61 alleles were obtained from 15 SPF Yorkshire,22 SPF Landrace and 330 SPF piglets.And there were 43 alleles of SLA class ? and 18 alleles of SLA class ? in these 61 alleles.The results showed that one newly discovered alleles were SLA-3*05:03:02,New alleles were submitted to the ISAG Nomenclature Committee and got official names,and 17 temporarily named alleles have been gained official names.3?Based on the polymorphism of SLA gene,we found that the polymorphism of SLA class I allele was mainly concentrated in exon 2 and exon 3,and the polymorphism of SLA class ? was mainly concentrated in exon 2.The more popular alleles of SPF Yorkshire are SLA-1*08:11,SLA-2*04:01,SLA-3*05:03:02,SLA-DQB1 *04:01:01,SLA-DRB1*02:01:01,and SPF Landrace are SLA-1*15:01,SLA-2*02:02,SLA-3*04:02,SLA-DQB1*07:01:01,SLA-DRB1*06:01.The 61 SLA alleles were distributed in multiple populations,which indicated that SPF Yorkshire and Landrace have close relationship with Meishan pigs,CanadianYorkshire,Hanford,Banna pigs,Korean native pigs,Yucatan and Sinclair.4?We obtained 19 SLA haplotypes through the results of SLA classification,which including 17 SLA class ? haplotypes and 11 SLA class ? haplotypes.Among the 19 SLA haplotypes,3 SLA class ? haplotypes,Hp-37.0,63.0 and 73.0,were newly discovered and have not been obtained from other breeds.SPF Yorkshire has 8 specific haplotypes,which including 1 recombinant haplotype,and 2 shared haplotypes(Hp-35.23 and Hp-26 b.23)were found in SPF Yorkshire and Landrace.SPF Landrace has 12 specific haplotypes,which containing 2 recombinant haplotypes.All-mentioned haplotypes were obtained official name by ISAG SLA Committee.
